  • Effect of Acid and Alkali Stress on Seed Germination and Seedling Growth of Caragana versicolor

    With seeds of Caragana versicolor in Purang County, Tibet as the materials, the seed germination and seedling growth of C. versicolor were measured under acid and alkali stress, and the soil acidity and alkalinity for normal growth and development of C. versicolor were determined,so as to provide the theoretical basis for cultivation of C. versicolor in acidified or alkaline soil. The results showed that the seed germination of C. versicolor was promoted when treated by strong acid(pH 3) and strong alkali(pH 11) solutions, and the seedling effect was good. The results also indicated that C. versicolor was a kind of plant with strong acid and alkali resistance, suitable for cultivation in acid and alkali soil areas.

  • Slaughtering Performance and Meat Quality of Hainan Black Goat and Its Hybrid Offsprings with Nubian Black Goat

    In order to compare the slaughter performance and meat quality of Hainan black goats and hybrid offsprings of Nubian Black Goat and Hainan Black Goat, six 7-month-old Hainan black goats and six hybrid offsprings were selected to determine slaughter performance and muscle quality. The results showed that hybridization of Nubian black goat and Hainan black goat significantly improved the production performance and slaughter performance of hybrid offsprings(P>0.05). The meat quality of F1 hybrid black goat had no significant difference with that of Hainan black goat except for the hardness and elasticity(P<0.05). Therefore, the crossbred progenies of Nubian black goat and Hainan black goat have advantages in meat yield and meat performance, and the main meat quality indexes are not significantly different, which meets the market require-ments for high-yield and high-quality hybrid combinations, and can be used for cross promotion.

  • Diagnosis and Control of Classical PRRSV Strain Infection in a Pig Farm

    The suspected cases of PRRSV in a pig farm of Shandong Province were conducted pathologic autopsy, RT-PCR amplification, and sequencing and analysis of PRRSV N protein gene. The results showed that the homology of the nucleotide sequences of protein N was 99.7% be-tween the isolate and the classical strains(VR2332 and SD1), and 93.4% between the isolate and the highly pathogenic strains(JXA1, HuN4, SX-1 and TJM). The results suggested that the virus was classical PRRSV in the pig farm. The research would provide a reference for the diagnosis and targeted therapy of classical PRRSV infection in swine production.

  • Protective Effect of Bacillus subtilis Peptidoglycan (PG) on β-conglycinin-induced Intestinal Epithelial Cells Damage of Juvenile Carp (Cyprinus carpio)

    [Objective] The paper was to investigate the protective effects of different concentrations of Bacillus subtilis peptidoglycan(PG) on β-conglycinin-induced inflammatory injury in intestinal epithelial cells of juvenile carp(Cyprinus carpio). [Method] In 24-cell microplates, the intestinal epithelial cells(IECs) of juvenile carp were primarily cultured for 72 h at 26°C and 6% CO2, and then the IECs were randomly divided into6 groups with 4 replicates per group. One of the six groups was set as negative control group, and the other groups were all supplemented with 1.0 mg/mL β-conglycinin in culture medium to establish inflammatory injury. At 24 h post induction, the culture media were changed into B. subtilis PG culture media with the concentrations of 0(positive control group), 0.15, 0.30, 0.45 and 0.60 mg/mL, respectively. The samples were collected to measure the antioxidant and anti-inflammatory indices at 12, 24 and 36 h post culture. [Result] β-conglycinin exposure significantly decreased the activity of ASA, AHR, SOD, CAT, GPx, and increased the PC content and the mRNA expression of inflammatory cytokines(IL-1β, IL-8, TNF-α1,IL-10 and TGF-β). At 12, 24 and 36 h post PG treatment, the activities of ASA, AHR, SOD, CAT, GPx and the content of PC in cells decreased in a dose-dependent manner; the mRNA levels of IL-1β, IL-8 and TNF-α1 were down-regulated and those of IL-10 and TGF-β were up-regulated.[Conclusion] Different concentrations of B. subtilis PG could protect IECs oxidative damage induced by β-conglycinin and improve the antioxidant capacity of IECs. High concentration of PG could improve the anti-inflammatory ability of IECs by inhibiting inflammatory factors and promoting the gene expression of anti-inflammatory cytokines.

  • Developmental Changes of GHR Gene Expression in Multiple Tissues of Mink

    [Objective] The paper was to explore the changes of GHR gene expression in different tissues of mink. [Method] With American mink as the research object, the expression volumes of GHR gene in heart, liver, spleen, lung, kidney, intestine and skin tissues at different growth stages(45, 90, 120, 150 and 180 days of age) were detected by real-time PCR, and comparative analysis was performed. [Result] The GHR gene expressions in heart, liver and spleen tissues at 180 days of age were extremely higher than those at other days of age( P<0.01). The GHR gene expression in lung tissue at 120 days of age was extremely higher than those at other days of age( P<0.01). The GHR gene expressions in intestine tissue at 45 and 120 days of age were extremely higher than those at other days of age(P<0.01), but no significant difference was observed between 45 and 120 days of age(P>0.05). The GHR gene expression in kidney tissue at 150 days of age was significantly higher than those at other days of age( P<0.05).The GHR gene expression in skin tissue was extremely higher than that those at other days of age( P<0.01). The GHR gene expressions in intestinal tissue at 45 and 120 days of age were extremely higher than those in other tissues(P<0.01). The GHR gene expressions in skin tissue at 90 days of age was extremely higher than those in other tissues(P<0.01). The GHR gene expressions in intestine, spleen and kidney tissues at 150 days of age were extremely higher than those in other tissues(P<0.01). The GHR gene expressions in spleen tissue at 180 days of age was extremely higher than those in other tissues(P<0.01). [Conclusion] The expression of GHR gene in mink showed obvious spatio-temporal specificity.

  • Effects of "Xinsanxian" on Growth Performance of Piglets and Immune Effect of Swine Fever Vaccine

    [Objective] The paper was to study mixed feed additives that could prevent and control diarrhea, improve piglet production performance and immune function, and substitute for antibiotics. [Method] Totally 100 healthy weaning piglets with an average age of 21 days were divided into five groups(half male and half female, similar weight), and additives were mixed in feed. Group A was added with Xinsanxian I composed of Xin-changtai, acidifier, suckling pig enzyme, etc.; group B was added with Xinsanxian II composed of Xinchangtai, Bacillus subtilis, yeast, etc.; group C was added with Xinsanxian III composed of Xinchangtai, fructooligosaccharide, suckling pig enzyme, etc.; group D was added with Jiaosanxian composed of hawthorn, malt, medicated leaven, etc.; group E was set as blank control group. Feed intake was recorded in columns throughout the exper-iment. Fasting piglets were weighed at the beginning(28 days of age) and the end(70 days of age) of the test, and the average daily weight gain(ADG), average daily feed intake(ADFI), feed gain ratio(F/G) and diarrhea rate of each piglet were calculated. Five piglets were randomly selected from each group before immunization, at 2 weeks post immunization, at 4 weeks post immunization and at 6 weeks post immunization, respectively.Approximately 2 mL of blood samples were collected from anterior vena cava, and the content of swine fever antibody was determined by ELISA.[Result] In terms of improving the growth performance of weaning piglets and preventing and controlling diarrhea, groups A and C had significantly better effects than groups B and D, all of which had significantly better effects than group E. The antibody titers of swine fever in group B and group C were significantly higher than those in group A and group D, all of which were significantly higher than that in group E( P<0.05). [Conclusion]Addition of Xinsanxian in the diet enhanced the immune effect of piglet swine fever vaccine, and improved the antibody titer. Moreover, it signifi-cantly increased the daily gain of weaning piglets, and reduced weight gain ratio and diarrhea rate. Xinsanxian III received the best effort.

  • In Vitro Probiotic Effect of Pseudostellaria heterophylla Fibrous Roots on Feed Bacillus subtilis and Preliminary Study on Its Solid Fermentation

    Traditional Chinese medicine Pseudostellaria heterophylla fibrous roots have been widely studied and applied in non-antibiotic clinical breeding of livestock and poultry. In vitro probiotic effect of P. heterophylla fibrous roots extract on feed probiotics Bacillus subtilis BD-K010 was studied, and the feasibility of solid fermentation of P. heterophylla fibrous roots was preliminarily evaluated. For in vitro probiotic effect, the increased concentration of P. heterophylla fibrous roots extract dependently increased the total biomass of B. subtilis BD-K010; 1.0% P. heterophylla fibrous roots extract received the best effect, and the final p H of 1.0% experimental group was closer to neutral. Meantime, B. subtilis BD-K010 with optimum concentration of P. heterophylla fibrous roots showed significantly higher in vitro antibacterial effect than the control group(P<0.01),and the antibacterial effects on Escherichia coli and Staphylococcus aureus were improved by 51.99% and 63.16%, but it was ineffective to Salmonella. For solid fermentation, the profile of substrate complex and appendage flocculent structure on substrate surface at the end of fermenta-tion in experimental groups added with B. subtilis BD-K010 and cellulase plus BD-K010 were more complex; the live bacteria number, polysaccharide content and saponin content at fermentation end-point in two experimental groups were extremely higher than those in the control group(P<0.01).P. heterophylla fibrous roots extract had good in vitro probiotic effect on B. subtilis BD-K010 and promoted its antibacterial effect, and it is feasible to use probiotics for solid fermentation of P. heterophylla fibrous roots and to improve effective components. It is of great significance to further de-velop and utilize P. heterophylla fibrous roots resources in modern animal husbandry.

  • Biological Characteristics of Lepus timibus in Captivity

    The biological characteristics of Lepus timibus in captivity, such as living habit, feeding habit, nutrition composition of main food, growth and development, molting rule, were studied systematically. The results showed that there were more than 30 kinds of common plants favored by L.timibus. The linear regression equation of weight gain was y=-23.223+30.929 x. The molting sequence in spring successively was head, dorsum, ear,both sides of the chest, abdomen, limbs, tails.

  • Histological Observation of Gonads of Male Elk (Elaphurus davidianus)

    Elk (Elaphurus davidianus) is a rare species native to China, and much less efforts have been dedicated to gonadal histology of male elk.The microscopic morphology of gonadal tissue of elk was observed by paraffin method. The results showed that the tunica albuginea testis was com-posed of collagenous fibers and elastic fibers, with the thickness of about 1 054 μm, and a large number of blood vessels were distributed in tunica albuginea; the seminiferous tubule was about 203 μm in diameter, with uneven thickness of tubule wall, which was consisted of sustentacular cells and spermatogenic cells; there were interstitial cells and blood vessels between seminiferous tubules. The epididymis contained more than 20 effer-ent ducts and a long curly epididymal duct; the capsule thickness at epididymis head was about 53 μm, and that at epididymis body and tail was400-1 000 μm; the epithelium of efferent duct was pseudostratified columnar epithelial cells; the wall epithelium of epididymal duct was composed of tall columnar cells at proximal lumina and stroma cells at distal lumina; the epithelial basement membrane of efferent duct and epididymal duct contained circular smooth muscle. The wall of spermatic duct was divided into mucosae, tunica muscularis and tunica adventitia. The tunica muscu-laris was well developed, which was smooth muscle fiber.
