  • Thickness Method for Measuring Hair Density of Rex Rabbit

    [Objective] The paper was to characterize hair density by changes in hair thickness under certain pressure.[Method] The effects of weight of thickness tester and wafer area at the bottom of presser foot on measurement results were studied,and the optimal measuring weight and wafer diameter was determined.The measurement results of hair thickness under the above condition were compared with the results of manual counting.Meanwhile,the effects of hair length and hair fineness on hair density were analyzed.[Result] The hair density obtained by manual counting had consistent change trend with the hair thickness measured under certain conditions.If the length and fineness of hairs were introduced for modification,the correlation between hair thickness and hair density would be better.There were little changes in hair length and fineness of rex rabbit,and their effects on hair thickness could be ignored under general condition.[Conclusion] It is completely feasible to characterize hair density of rex rabbit by hair thickness under pressure.

  • Effect of Fermentation Bed with Spent Mushroom Substrate of Pleurotus eryngii on Weaning Piglets

    [Objective] The paper was to investigate effects on fermentation bed temperature,growth performance,diarrhea rate and digestive en-zyme activity of weaning piglets by using spent mushroom substrate of Pleurotus eryngii as padding.[Method] A total of 120 weaning piglets(Duroc × Landrace ×Yorkshire) with average initial body weight of(8.0 ±0.5)kg were allocated to five dietary treatments in a randomized complete block design for 42 d,each of which was replicated three times with eight piglets per replicate(half male,half female).The padding for control group was(50% sawdust +50% rice husk);experimental group Ⅰ 100% spent mushroom substrate;experimental group Ⅱ(15% sawdust +15% rice husk +70% spent mushroom substrate);experimental group Ⅲ(25% sawdust +25% rice husk +50% spent mushroom substrate);experimental group Ⅳ(35% sawdust +35% rice husk +30% spent substrate).[Result] There was no significant difference in surface temperature of fermentation bed between experimental groups and control group(P>0.05).Compared with the control group,the temperature under 20 cm of fermentation bed in ex-perimental groups I,Ⅱ and Ⅲ increased significantly(P<0.05).Except for experimental group Ⅳ,other three experimental groups had higher aver-age daily gain(P<0.05) and experimental group Ⅰ had higher average daily feed intake(P<0.05) compared to the control group.The diarrhea rate and mortality of weaning piglets in experimental groups Ⅱ,Ⅲ and Ⅳ were significantly decreased compared with the control group(P<0.05).Compared with the control group,other three experimental groups had higher digestive enzyme activity in duodenal contents except for experimental group Ⅳ(P<0.05).[Conclusion] Spent mushroom substrate of P.eryngii can be used as fermentation bed padding,and the optimal proportion was experimental group Ⅲ.

  • Changes in Chilled Chicken Quality and Correlation Analysis during Different Shelf Life

    [Objective] The paper was to study the change rule and correlation analysis of quality indexes of chilled chicken within 7 d shelf life.[Method] Six pieces of fresh breast muscle were packaged in polyethylene(PE) food storage bags,labeled,and refrigerated at 4 ℃.The p H,water loss rate,tenderness,color value and inosinic acid content of chicken were measured at different time points of preservation(cold storage for 0,1,2,3,6,7 d).[Result] With the prolongation of shelf life,the p H of chilled chicken first increased then tended to be stable;the water loss rate first increased then stabilized;the shear force first decreased then increased.There was little difference in brightness value(L*) of chilled chicken dur-ing different shelf life(P>0.05).The red degree value(a*) decreased with the prolongation of shelf life;the yellow degree value(b*) increased with the extension of shelf life.The inosinic acid content decreased with the prolongation of shelf life.The p H of chilled chicken in different shelf life had significant positive correlation with water loss rate(P<0.05) and extremely significant negative correlation with shear force(P<0.01).[Conclusion]The shelf life of 7 d has impact on meat color and inosinic acid of chilled chicken.Cold fresh preservation of chilled chicken should be less than 6 d;p H has certain correlation with water loss rate and shear force.

    In recent decade,the fluctuation of pig price has become more and more acute with obvious periodicity.Currently,the pig breeding mode dominated by individual households leads to acute fluctuation of pig price to some extent.The scale expansion of recent decade intensified the fluc-tuation of pig price in the short term but will stabilize the price in the long term.Therefore,it is necessary to expand breeding scale for maintaining long-term price stability and the optimal scale should be chosen according to the circumstances to ensure the social benefit maximization.

  • Ozone Disinfection Test in Chicken House

    [Objective] The paper was to verify the disinfection effect of ozone on livestock and poultry houses.[Method] The empty chicken house was disinfected by ozone with the help of ozone disinfector.[Result] As the ozone disinfector was worked for 60 min,the average sterilizing rate of air was 85.33%;the average sterilizing rate on the surface of chicken coop was 86.77%;and the average sterilizing rate of wall was 89.13%.The disinfection effect was obvious.[Conclusion] The results will provide references for promotion and application of ozone disinfector in breeding in-dustry.

  • Isolation and Identification of Photobacterium damselae subsp.damselae(PDD) from Tongue Sole

    In May 2016,an epizootic occured among cultured tongue soles caused mass deaths in a fish farm in Qinhuangdao,China.In order to find out the etiological agent,a bacterial strain was isolated from ascites and other tissues of sick tongue sole aseptically collected.The isolate was identified as Photobacterium damselae subsp.damsela(PDD) by isolation culture,Gram staining,physiological identification,morohological observation,biochemical identification and 16S rDNA sequence analysis.The results showed that the isolate shared 99.6% homology with the reference strain in GenBank.The animal regression test displayed that the isolate had very strong pathogenicity to tongue sole.The LD_(50) was 3.1 × 10~4 CFU/mL,and it showed pathogenicity to mammals.The antimicrobial susceptibility test showed the isolate was highly sensitive to nrofloxacin,Norfloxacin,Ciprofloxacin,Mequindox;moderately sensitive to Cefradine,Doxycycline;and insensitive to Gentamicin,Ceftriaxone,Tilmicosin,etc..

  • DNA Barcoding:An Effective Tool for Species Identification of Lepidopteran Oak Pests

    [Objective] The paper was to improve the efficiency and accuracy of early forecast of Lepidopteran oak-infesting pests.[Method] DNA barcoding technique was established for quick species identification using mitochondrial cytochrome C oxidase subunit Ⅰ(COⅠ) as the standard gene.This barcoding technique was used to amplify and sequence genomic DNA samples from eggs and pupae of 11 species of Lepidopteran pests collected from oak.[Result] The DNA barcoding standard genes of 594-708 bp were determined from eggs and pupae of Lepidopteran insects.There were differences of 0-2 bases in DNA barcode sequences between conspecific eggs and pupae,with the sequence identity of 99.7%-100%.The average content of A,T,G and C of DNA barcode sequences from Lepidopteran insects were 30.7%,38.5%,14.9% and 15.9%,respectively.The obtained DNA barcode sequences had 91.4%-100% identity and 0-8.6% difference degree with GenBank-deposited DNA barcode sequences from organisms of the genetically-closest relationship.Among them,DNA barcode sequences from egg and pupa samples of 10 Lepidopteran insects(No.1-20) had 99%-100% identity and 0-1.0% difference degree with homologous sequences in GenBank database,while the remaining samples(No.21-22) had high difference degree(8.6%) with homologous sequences.[Conclusion] The established DNA barcoding technique is an effeetive tool for species identification of Lepidopteran pests using genomic DNA from eggs and pupae of Lepidopteran insects.

  • Effects on Growth Performance and Feed Digestibility of Hainan Black Pigs by Adding Lysine in Low Protein Diet

    [Objective] The paper was to study the effects on growth performance and feed digestibility of Hainan black pigs by adding lysine in low protein diet.[Method] A total of 36 Hainan black pigs with the body weight of(46.34 ±3.06) kg were randomly divided into three groups:control group(protein level 15%,lysine level 0.83%),experimental group I(protein level 13%,0.83%),and experimental group II(protein level 13%,lysine level 1.03),each group had three replicates of four pigs.The trial lasted 35 d.[Result] The growth performance of Hainan black pig in group II had no significant difference with that in control group(P>0.05).The apparent digestibility of crude protein and phosphorus of Hainan black pigs in Group II increased significantly(P<0.05),which were increased by 10.08% and 11.85% respectively compared to the control group;the apparent digestibility of calcium was increased,but the difference was not significant(P>0.05).[Conclusion] It will not reduce the growth performance of Hainan black pig when the protein level of diet is reduced by 2% and the lysine level is increased to 1.03%.It is recommended that the diet can be used in the production of Hainan black pig.

  • Effect of N-Carbamylglutamate(NCG) on Growth Performance of ICR Mice

    To understand the effects of self synthesized N-carbamylglutamate(NCG) on growth performance of ICR mice,mice were fed with the basal diet adding with 0.025%,0.05% and 0.1% NCG for 28 d.Effects of different levels of NCG on body weight of mice were observed.The re-sults showed that the addition of NCG in the feed could improve the growth performance of mice,and the growth-promoting effect enhanced with the increasing dose.

  • Construction and Immunogenicity of Recombinant Lactococcus lactis Expressing S1 Protein of Porcine Epidemic Diarrhea Virus(PEDV)

    To evaluate the specific immune responses induced by recombinant Lactococcus lactis(L.lactis) which expresses porcine epidemic diarrhea virus(PEDV) S1 protein through oral administration,the spike gene fragment of PEDV was amplified from PEDV SDLY strain to construct p MG36 e-S1 recombinant plasmid.The recombinant plasmid was then electro-transferred into competent cells of L.lactis MG1363,to prepare the recombinant L.lactis expressing S1 protein of PEDV.The expression of target protein was identified by SDS-PAGE and Western-blot.New Zealand white rabbits were orally administered with the recombinant strain;the antibody titer in intestinal mucosa and serum was detected by neutralizing test;and the specific Ig G in serum was evaluated by indirect ELISA.The results showed that the recombinant L.lactis could effectively induce high level of Ig G in serum and high level of mucosal immune antibody.The recombinant L.lactis is qualified to be a potential oral vaccine because it could successfully stimulate both humoral and mucosal immune responses against PEDV.

  • Isolation and Identification of Sheep Pseudorabies Virus

    In a sheep farm with mixed culture of pig and sheep in Shandong Province,sheep were attacked by a disease featured by foaming at the mouth,neurological symptoms and partial hair slip of legs,and the mortality of the disease was as high as 100%.In order to determine the pathogen,dead sheep were analyzed through pathogen isolation,PCR assay and direct immunofluorescence identification,and the pathogen was confirmed as pseudorabies virus(PRV).Sequencing results showed that the g E gene of the isolated strain shared the homology of 97%-99% with the nucleotide sequence of known PRV genome in the NCBI databases,suggesting the isolate was PRV.The virus had obvious cytopathic effect through BHK cell line passage till the seventh generation,and the amount of half virus tissue cell infection(TCID50) was 1×107.5/m L following ReedMuench method.Two healthy sheep with the body weight of 20 kg were injected with the viral fluid of the isolate,and typical symptoms of pseu-dorabies(PR) were observed after 4 d.According to clinical symptoms and PCR diagnosis results,the epidemic situation of sheep farm was effec-tively controlled through comprehensive measures such as eliminating swinery in the farm,strengthening disinfection of pigsty,injecting sick sheep with pseudorabies serum,supplementing healthy sheep herb with antivirus traditional medicine Qiqing Baidu granule.

  • In Vitro Antibacterial Test of Alkaline Hydrogen Water on Four Kinds of Common Pathogenic Microorganisms

    [Objective] The paper was to investigate in vitro antibacterial effect of alkaline hydrogen water on Escherichia coli,Salmonella,Pseudomonas aeruginosa and Staphylococcus aureus.[Method] With Cortex cinnamomi extract(1 g/m L) and pure water as the control,the minimal inhibitory concentrations(MIC) against four kinds of common pathogenic microorganisms were tested through microdilution method.[Result] When the alkaline hydrogen water was diluted to 1/8 times of the original concentration,it had significant antibacterial effects on four kinds of common mi-croorganisms with the concentration of 1.5 ×10~5 CFU/m L,which had equivalent effect with C.cinnamomi extract group.[Conclusion]The alkaline hydrogen water has remarkable antibacterial effects on the four kinds of common microorganisms,which may provide a new important way for pre-venting disease occurrence and reducing the harms of pathogenic microorganisms.

  • Isolation and Identification of Haemophilus parasuis SD02 strain

    A bacterial strain was isolated from the sick pigs suspiciously infected by polyserositis and arthritis in a pig farm in Shandong Province,and identified through morphological observation,culture traits,biochemical characteristics and PCR amplification.Additionally,primers were de-signed according to the 16S rRNA sequence of Haemophilus parasuis,and the bacterial strain was amplified by PCR.The amplified fragments of approximately 1 400 bp was sequenced,and aligned with the sequence in Gen Bank.The results showed that it shared the homology of 97%-99%with the 16S rRNA sequence of foreign H.parasuis,and confirmed as H.parasuis(HPS).The strain was determined as serotype 4 through serotype identification.The strain was named SD02.

  • Diagnosis and Treatment Report of the First Case of Mycoplasma bovis in Shandong Province

    [Objective] The paper was to introduce the first case of Mycoplasma bovis in Shandong Province.[Method] Samples were collected from sick cattle,and identified through M.bovis isolation,biochemical identification,PCR,16S RNA sequence analysis and clinical treatment and other research work.[Result] A strain of M.bovis was successfully isolated,and typical colonies in fried egg shape were grown.It was further confirmed as M.bovis via molecular biological detection.The use of vaccine and sensitive drugs in clinical could quickly control the epidemic situation of cat-tle herds.[Conclusion] This is the first reported case of M.bovis in Shandong Province,and its diagnosis and treatment process is conducive to further prevention and control of the disease.

  • Effect of Ferulic Sesquiterpene Compounds on Enzyme Activity of Schistosomulum

    [Objective] The paper was clear the effects of ferulic sesquiterpene compounds on enzyme activity of schistosomulum.[Method]Mice infected with schistosome cercariae were orally administrated with ferulic sesquiterpene compounds at the dose of 300 mg/kg on the 6 thday post infection once a week for 4 weeks.Schistosomula were collected and ground into homogenate on the 3 rdand 5 thday after every time of administration.Thin layer starch gel electrophoresis was used to observe the activity of enzymes in the homogenate.Control group was also designed(drug free mice).[Result] The activities of malic dehydrogenase,6-phophate mannosease and acid phosphatase were obviously inhibited in the drug group,but the activity of phosphoglucoisomerase was not inhibited.[Conclusion] Ferulic sesquiterpene compounds have inhibitory effect on energy metabolism of schistosomulum and digestion of red blood cells.

  • Screening of Traditional Chinese Medicine Prescription for Treating Damp-heat Diarrhea of Piglets

    [Objective] The paper was to explore the therapeutic effect of traditional Chinese medicine on damp-heat diarrhea of piglets in summer.[Method] Three prescriptions were developed with a dozen of traditional Chinese medicines,and in vitro antibacterial test against swine standard Escherichia coli [O149:K91,K88 ac(C83907)] was conducted.Moreover,106 natural cases of diarrhea piglets caused by coinfection of damp-heat and E.coli through clinical and laboratory diagnosis were then treated.[Result] The oral liquids prepared by three traditional Chinese medicine prescriptions had certain antibacterial effect on standard pathogenic E.coli.Prescription 2 presented the strongest inhibitory effects on swine E.coli in vitro,with the MIC of 31.25 mg/m L.It had better therapeutic effect,and the effective rate reached 94.74%.[Conclusion] The prescription 2 mainly consisted of Coptis chinensis and Prunus mume might be effective in treatment of piglet diarrhea caused by damp heat and E.coli in clinic.

  • Phylogenetic Analysis of HN Gene of Eight Pigeon NDV Isolates in Guangxi

    [Objective] The paper was to provide a basis for scientific prevention and control of pigeon Newcastle disease(ND).[Method] The HN gene of eight pigeon NDV strains isolated from different pigeon farms in Guangxi were amplified by RT-PCR,sequenced and analyzed.The molecular evolution characteristics of HN gene of pigeon NDV isolates in Guangxi was discussed.[Result] The nucleotide sequence length of HN gene of the eight NDV isolates was 1 716 bp,encoding 571 amino acids.They belonged to virulent group C,and the gene length characteristic of HN gene accorded with virulent strain.Analysis of nucleotide homologies indicated that the eight NDV isolates shared higher homology with genotype VIb,ranging from 90.4% to 99.5%.Phylogenetic tree analysis demonstrated that the genetic relationship between the eight NDV strains in Gangxi and the NDV isolates from Guangxi,Guangdong,Jilin,Liaoning,Yunnan and Heilongjiang during 2011 and 2013 was close.They were located in the same cladogram branch.[Conclusion] We assume that the eight pigeon NDV isolates in Guangxi all belong to the gene class II genotype VI b NDV.

  • Transplantation Therapy of Cats with Chronic Renal Failure

    Dialysis therapy is not a radical cure of chronic renal failure in cats,while renal transplantation is the cure method.Renal transplanta-tion requires rigorous examination of donor and receptor,and the procedure is strictly sterile.The main points of renal transplantation are:renal artery and vein sutured to abdominal aorta and posterior vena cava,respectively;ureter sutured to bladder;minimizing the time of thermal ischemia.Cats should be given immunosuppressant before the operation,and the drug should be administered throughout the life.Various indicators should be monitored regularly to prevent the occurrence of immunological rejection.

  • Detection of Patulin in Dairy Products by ELISA

    The content of patulin in dairy products was detected by rapid ELISA assay.The pretreatment of samples was simple,with wide range of application.The standard product showed good linear relationship with the range of 0-5.4 μg/kg,and the linear relationship reached 0.999 3,with high sensitivity.It took shorter time,with a total time-consumption of 1 h.Moreover,it had good repeatability for analytical requirements.
