  • Experimental Analysis on Allometric Growth in Spualiobarbus curriculus Larvae and Juveniles in Zhaoqing Reach of the Pearl River

    Ecological experiment was conducted in this paper to study the allometric growth in Squaliobarbus curriculus larvae and juveniles from Zhaoqing Reach of the Pearl River. The results showed that after first feeding, the organs associated to swimming, feeling and feeding of larvae differentiated rapidly, and exhibited allometric growth. The head length, tail length and head height exhibited positive allometric growth, while trunk length exhibited negative allometric growth before reaching the growth inflexion point 20 d after first feeding, and positive allometric growth after the inflection point. Body height exhibited positive allometric growth both before and after the growth inflexion point at 18 d after first feeding. The inflexion points of snout length, eye diameter and post-eye head length occurred 23, 19 and 16 d after first feeding. For swimming organs, the dorsal fin, pelvic fins and anal fin exhibited positive allometric growth both before and after the inflexion point at 18, 19 and 17 d after first feeding; but the tail fin exhibited positive allometric growth before the inflexion point at 14 d after first feeding, and negative allometric growth after the inflexion point. S. curriculus larvae can possess various abilities soon after first feeding to survive at early stage due to the rapid development of swimming, feeling, feeding and other organs, which has important ecological significance to adapt to the complex external environment.

  • Effect of Secondary Sterilization on the Quality of Roasted Whiteleg Shrimp(Litopenaeus vannamei)

    This study was carried out to investigate the effects of secondary sterilization on the quality of roasted shrimp( Litopenaeus vannamei)with high nutrients. Roasted shrimps were vacuum packaged, with 100 g in each bag, and subjected to secondary sterilization for 30 min at 70, 80,90, 100, 110 and 120 ℃, or for 10, 20, 30, 40 and 50 min at 90℃. Then, the color, textural properties, sensory quality and pH were measured, and whether they were commercially sterile was evaluated. The results showed that with the increase of sterilizing temperature, L*, a*and b*values all decreased. And with the increase of the duration of sterilization, L*slightly decreased, while a*and b*slightly increased. In addition, with the increase in sterilizing temperature, the hardness, gumminess, chewiness and springiness of roasted shrimps increased at first and decreased subsequently, while the adhesiveness kept increasing, and the cohesiveness and shear force slightly declined. With the increasing duration of sterilization,the hardness, adhesiveness and chewiness rose, the cohesiveness and shear force declined, the gumminess decreased at first and increased subsequently, while the springiness increased at first and decreased subsequently. The pH of roasted shrimps varied in a complex pattern when sterilizing temperature was increased, but increased with the increasing duration of sterilization. The results showed that the roasted shrimps secondarily sterilized at 90℃ for 30 min have the best sensory quality, and are commercially sterile.

  • The Genetic Structure and Diversity of Repomucenus curvicornis Inhabiting Liaoning Coast Based on Mitochondrial COⅠ Gene and Control Region

    [Object] This study was conducted to explore the genetic diversity and structure of the wild Repomucenus curvicornis inhabiting Liaoning Coast, China. [Method] The mitochondrial COⅠ gene and control region(CR) were PCR amplified from the wild R. curvicornis populations from the Liaodong Bay(n=22) and the northern Yellow Sea(n=18), sequenced and analyzed for genetic diversity. [Result] The contents of A, T, C and G of 624 bp COⅠ gene were 24.09%, 31.04%, 25.28%, and 19.59%, and those of 460 bp CR fragment were 32.96%, 32.80%, 14.86% and 19.38%, respectively. The total number of variable sites, average number of nucleotide differences( k), haplotype diversity(H) and nucleotide diversity(π) based on COⅠ gene were 38, 4.67,(0.96±0.02) and(0.007 5±0.004 2), and those based on CR fragment were 26, 3.35,(0.97 ±0.02) and(0.007 3±0.004 3), respectively. Based on mitochondrial COⅠ gene and CR, the genetic diversity of Liaodong Bay population was lower than that of the northern Yellow Sea population. The AMOVA analysis based on CR fragments revealed almost significant genetic divergence between the Liaodong Bay and the northern Yellow Sea populations, while there was no significant genetic divergence based on COⅠ gene. The results showed that CR and COⅠ gene are effective molecular markers for detecting the genetic diversity of R. curvicornis population, while CR is more reliable than COⅠ gene in detecting the genetic structure. [Conclusion] CR is an appropriate marker for genetic analysis of marine fish population.

  • Effect of Poly-β-Hydroxybutyrate on the Activity of Nonspecific Immunity Related Enzymes in Fenneropenaeus chinensis

    [Objective] This study was carried out to explore the effect of poly-β-hydroxybutyrate(PHB) on the performance of the nonspecific immune system in Fenneropenaeus chinensis. [Method] F. chinensis individuals were assigned into six groups, and each group was fed with a diet containing 0(Control), 0.5%(Group E_(0.5)), 1.0%(Group E_(1.0)), 2.5%(Group E_(2.5)), 5.0%(Group E_(5.0)) or 10.0% PHB(Group E_(10.0)). The mortality rate and relative percent of survival(RPS) of each group were calculated after 6 weeks. Meanwhile, the total antioxidant capacity(T-AOC), the activity of acid phosphatase(ACP), peroxidase(POD) and catalase(CAT), and the content of malondialdehyde(MDA) in hepatopancreas and serum were measured,and their correlation with PHB concentration was analyzed. [Result] The RPS in PHB treated shrimps increased initially and decreased subsequently with increasing PHB concentration. RPS of Group E_(1.0) was the highest, showing significant difference from that of other groups( P <0.05).With the increase in PHB concentration, the activity of immunity related enzymes changed in a similar pattern with RPS: increasing at first and decreasing subsequently. In addition, the activity of the enzymes was elevated in the 2~(nd) and 3~(rd) weeks of PHB administration. Among them, T-AOC in serum of groups E_(1.0) and E_(2.5), T-AOC in hepatopancreas of Group E_(1.0), ACP activity in serum of groups E_(1.0) and E_(2.5), ACP activity in hepatopancreas of Group E_(1.0), CAT activity in groups E_(0.5), E_(1.0) and E_(2.5), CAT activity in hepatopancreas of groups E_(0.5), E_(1.0) and E_(10.0), POD activity in both serum and hepatopancreas of groups E_(0.5), E_(1.0) and E_(2.5), SOD activity in both serum and hepatopancreas of Group E_(1.0), MDA content in serum of Group E_(1.0) and MDA content in hepatopancreas of groups E_(0.5) and E_(1.0) showed significant difference from those of other groups(P<0.05). [Conclusion] PHB can improve the immunity of F. chinensis, 1.0% in feed has the best effect, and the total enzyme activity reaches the highest level in the 2~(nd) and 3~(rd) weeks of PHB administration.

  • Study on Net Hauler Safety Device for Fishing Vessels

    [Objective] To design a safety device set for fishing vessel net hauler, in order to reduce the severe potential safety hazards in offshore operation. [Method] In view of the operation and structure of net hauler on the fishing trawler, this paper investigated the existing safety protection devices and designed a new set of purely mechanical structure without changing the original structures of the vessel significantly. [Result] Using the easily-adjustable functions of the main shaft of CNC lathe, and simulating the operation performance of different hauling line speeds and testing the safety device, the results showed that the new device could protect the crew from safety accidents caused by net hauler. [Conclusion] The result provides references for the design and application of mechanical protection devices for net haulers on other fishing trawlers.

  • Effects of Dietary Yeast Cell Wall Polysaccharides Supplementation to Late Gestation and Lactation Diets on Reproductive Performance and Blood Profiles of Sows and Their Offspring

    This study was conducted to determine the effects of yeast cell wall polysaccharides(YCWP) on reproductive performance, serum profiles and milk composition during late gestation and lactation. A total of 96 sows(Landrace × Yorkshire) were blocked on a basis of breed, parity(2-3)and their respective d 85 of gestation, and then randomly allotted to the following three dietary treatments: CON(basal diet, treatment 1), YCWP1(CON+0.2% YCWP, treatment 2) and YCWP2(CON+0.4% YCWP, treatment 3). The experiment lasted from d 85 of gestation to weaning(d 21 of lactation). Sows fed YCWP diets had lower backfat thickness reduction from postpartum to weaning( P<0.05). The supplementation of YCWP tended to increase litter weight at weaning(P=0.08) compared with CON. The non-lipid and total milk solids in colostrum was improved by YCWP2(P<0.05).No differences were observed in milk composition among dietary treatments( P>0.05). The supplementation of YCWP decreased(P<0.05) the levels of alanine amino transferase(ALT), aspartate amino transferase(AST), while it did not affect( P>0.05) alkaline phosphatase(ALP), total protein(TP),albumin(ALB), globulin(GLB), glucose(GLU), total cholesterol(TC), triglyceride(TG) or immunoglobulin G(IgG) in the sow serum. The serum IgG of piglets was increased by the YCWP. The levels of serum ALT and AST were reduced(P<0.05) for suckling piglets in YCWP treatments. Taken together, the results of current studies might indicate beneficial effects of YCWP supplementation to late gestation and lactation diets on the sow productivity by improving litter weight at weaning, reducing backfat loss, and increasing serum IgG concentrations of piglets.

  • Effects of Post-expanding Lower Temperature Granulating Process on Growth Performance of Piglets

    [Objective] The paper was to compare the effects of traditional granulating process and post-expanding lower temperature granulating process on starch gelatinization degree and in vitro protein digestibility of conversation feed, and growth performance of piglets. [Method] Sixty weaned piglets, 25 days old, were randomly divided into two groups, six replicates each group, and each replicate contained five piglets. Piglets in control group were fed with the conversation feed developed by traditional granulating process, and those in experimental group were fed with the conservation feed developed by post-expanding lower temperature granulating process. The trial lasted 28 d. [Result] Compared to traditional granulating process, post-expanding lower temperature granulating process significantly enhanced starch gelatinization degree of conservation feed by56.9%(P<0.05), and significantly improved in vitro protein digestibility of conservation feed by 11.48%(P<0.05). Compared to the control group, the feed intake of piglets in experimental group was increased by 21.63%(P<0.05); the average daily gain was increased by 27.61%(P<0.05); the diarrhea rate was lowered; and the feed gain ratio was improved by 4.51%(P>0.05). The results suggested that post-expanding lower temperature granulating process effectively enhanced feed digestion and absorption of piglets. [Conclusion] The paper provides a theoretical basis and practical reference for improvement of production process of conservation feed.

  • Herbaceous Cultivation Test of Mulberry in Sichuan Sericultural Area

    With Guiteyou 2 as the test mulberry variety, mulberry herbaceous cultivation field was established at the planting densities of 60 000,90 000 and 120 000 plants/hm~2, respectively. Hybrid mulberry herbaceous cultivation test was conducted, with Tongxiangqing mulberry field of mid-fist-form at conventional planting density of 4 995 plants/hm~2 as the control. The results showed that the leaf yield of herbaceous cultivation mulberry was closely related to the planting density. With the increasing planting density, the leaf yield per plant was decreasing, but the leaf yield per unit area of mulberry field was increasing. The annual leaf yield per unit area in the mulberry field with the average planting density of 120 000 plants/hm~2 was 37 560 kg/hm~2, 2.14 times of that of the control field. The moisture content of mulberry leaf in herbaceous cultivation field was 4.74% higher than the control; the contents of crude protein, crude fat, crude fiber and crude ash were 1.264%, 0.014%, 0.744% and 0.002%lower than the control, respectively. Comparison of leaf rearing with herbaceous cultivation mulberry and control mulberry showed that there was no significant difference in growth status of silkworm. The cocoon weight, cocoon shell weight, cocoon yield per 10 000 larvae, cocoon filament length and non-broken filament length of Chuanshan × Shushui were 12.81%, 14.29%, 13.85%, 5.95% and 7.68% lower than the control, respectively; but the percentage of common cocoons was 0.16% higher than the control; the cleanness and neatness were 0.2 and 1.25 points higher than the control;the filament size was 0.196 dtex lower than the control. The cocoon weight, cocoon shell weight, cocoon yield per 10 000 larvae, cocoon filament length and non-broken filament length of Jingsong × Haoyue were 11.06%, 10.20%, 11.53%, 9.46% and 9.03% lower than the control, respectively;but the percentage of common cocoons was 1.77% higher than the control; the cleanness and neatness were 0.12 and 2.50 points higher the control;and the filament size was 0.196 dtex lower than the control.

  • Moisture Content and Nutrient Composition of Alfalfa under Different Drying Methods

    [Objective] The paper was to study the effects of different drying methods on moisture content and nutrient composition of alfalfa.[Method] Alfalfa collected from the same piece of land in early flowering period were dried through three different drying methods, solar drying,outdoor drying in the sun and outdoor drying in the shade, and the effects of different drying methods on contents of moisture, crude protein, crude ash and neutral detergent fiber(NDF) were measured. [Result] The moisture evaporation rate of different drying methods over the same period successively were solar drying > outdoor drying in the sun > outdoor drying in the shade. The crude protein content of alfalfa was the highest through solar drying(P<0.05), followed by outdoor drying in the shade, and the crude protein content through outdoor drying in the sun was the lowest. Under the condition of different moisture contents, the crude protein content of dried samples through outdoor drying in the sun decreased by 6.86%-22.43% compared to solar drying, and that through outdoor drying in the shade reduced by 4.92%-17.20%. Under the same moisture content, the crude ash content successively were outdoor drying in the shade > outdoor drying in the sun > solar drying; the crude ash content of alfalfa through solar drying was 3.72%-19.22% lower than outdoor drying in the sun, and 8.12%-29.93% lower than outdoor drying in the shade. The NDF content of alfalfa through outdoor drying in the sun was the highest, significantly higher than the other two drying methods( P<0.05). The NDF content through solar drying was the lowest, 9.08%-27.25% lower than outdoor drying in the sun, and 4.84%-24.84% lower than outdoor drying in the shade. [Conclusion] The paper will provide some references for selection of appropriate alfalfa drying method.

  • The Adaptive Planting and the Prospect of Industrial Development of Quinoa in China

    Quinoa is the only single plant meeting the basic nutritional needs of the human body. It is the whole nutritious food with nutritional balance, wide adaptability, strong resistance and other characteristics. In this paper, the botanical characteristics and nutritional value of quinoa were systematically introduced, and the development of quinoa in China was reviewed. The problems of domestic quinoa industry, the shortage of germplasm resources, the lack of innovation and high-yielding cultivation techniques were analyzed. To provide theoretical references for quinoa planting and industrial development in China, the countermeasures to improve the investment of quinoa industry and promote the development of multi-form industry were put forward.

  • Effects of Dietary Picria felterrae Lour Extracts on Serum Biochemical Indexes and Immune Function of Guangxi Partridge Chickens

    [Objective] This trial was to investigate the effect of dietary Picria felterrae Lour extracts on serum biochemical indexes and immune function of Guangxi Partridge chickens. [Method] Totally 500 individuals of 1-day-old Guangxi Partridge chickens were randomly divided into five groups, five replicates each group and 20 chicks each replicate. Groups A, B and C were treatment groups supplemented with 0.70%, 0.35% and0.175% P. felterrae extracts in the basal diet, respectively; group D was a medical control group supplemented with 0.01% colistin sulfate premix in the basal diet; and group E was a control group with basal diet. Serum samples were collected from 10 chickens in each group at 21, 35 and 49 days of age, to analyze the biochemical and immune indexes. [Result] It had no significant impact on serum TP and GLU contents of chickens by adding different levels of P. felterrae extracts in the basal diet(P >0.05). However, at 21 and 35 days of age, the TBILI content of chickens in groups A, B and C were extremely lower or significantly lower than that in group E( P<0.01; P<0.05). At 35 days of age, the serum AKP activities in groups A, B and C were extremely higher than that in group E(P<0.01), and the serum GOT activities at 21 and 35 days of age in group C were significantly higher than that in group E(P<0.05). At 49 days of age, the serum Ig G content in groups A, B and C and the IL-2 content in group B and group C were significantly lower or extremely lower than that in group E( P<0.05; P<0.01); the T-AOC content in group B was significantly higher than that in group E(P<0.05); the GSH content in groups A, B and C were significantly higher than that in group E( P<0.05). Adding different levels of P. felterrae extracts in the diet significantly decreased the TBILI content, reduced the Ig G, IL-2 and IL-6 contents, and improved the AKP and GOT activities and the T-AOC and GSH contents of Guangxi Partridge chicken. [Conclusion] Adding 0.35% P. felterrae extracts in the basal diet received the best effort.

  • Prokaryotic Expression and Preparation of Polyclonal Antibody of HtrA Protein from Escherichia coli Causing Pneumonia in Fox

    To investigate the roles of HtrA protein in bacterial adaption to stresses, htra gene was cloned from an Escherichia coli strain isolated from silver fox, double digested, and ligated into the prokaryotic expression vector pET32a. Then, the recombinant plasmid pET32a-htra was transformed into E. coli BL21 competent cells. The over-expression of His-HtrA protein was induced by the addition of IPTG at a final concentration of 1 mmol/L, and verified by SDA-PAGE and Western blot. In addition, the protein was purified with Ni-NTA agarose, and used to immunize mice,and the polyclonal antibody specifically bound to E. coli HtrA protein. The results will provide a theoretical basis for studying the roles of HtrA protein and developing vaccines for fur-bearing animals against E. coli.

  • Prevention and Treatment Test of Piglet Diarrhea

    [Objective] The paper was to observe the clinical effects of compound Chinese traditional preparation Shenwudi oral liquid on prevention and treatment of piglet diarrhea, and to explore the diarrhea prevention and growth promotion effect of Shenwudi power on weaned piglets as the medical feed additive. [Method] Diarrhea piglets were randomly grouped, and the clinical prevention and treatment effects of Shenwudi oral liquid were observed. Meantime, the effects of Huangzhi oral liquid and levofloxacin mesylate and sodium chloride injection were determined and compared with that of Shenwudi oral liquid. Shenwudi powder was added into the specific feed of weaned piglets at the dose of 5 g/kg, to observe its diarrhea prevention and growth promotion effect on weaned piglets. Compound amoxicillin powder was added in drug control group at the dose of 0.3 g/kg; no drug was added in blank control group. [Result] The cure rate of sucking piglets by Shenwudi oral liquid was 88.57%, and its protection rate was 100%; the cure rate of weaned piglets by Shenwudi oral liquid was 90.00%, and the total effective rate was 95.00%; the protection rate of weaned piglet diarrhea by Shenwudi powder mixing feed was 100%, and the average daily feed intake on the 42~(nd) day was extremely higher than the blank control group(P<0.01). [Conclusion] Shenwudi oral liquid can be used for prevention and treatment of sucking piglet diarrhea.Meantime, Shenwudi mixing feed has good prevention effect and significant growth promotion effect on weaned piglet diarrhea.

  • Isolation and Identification of Pathogenic Bacteria Causing Bovine Subclinical Mastitis in Eastern Hebei Province

    In this study, the milk samples of 1 021 cows in eight dairy farms in Eastern Hebei Province were collected and detected with LMT reagent and somatic cell count for subclinical mastitis. Pathogenic bacteria in subclinical mastitis positive milk samples were isolated and identified.The results showed that 60.63%(619/1 021) of the sampled cows were diagnosed with subclinical mastitis, and mixed infections accounted for 88.21%(546/619) of the cases. In addition, 82 strains of 14 species were isolated from the subclinical mastitis positive milk samples, including 36 strains of Staphylococcus(43.90%), 33 strains of Streptococcus(40.24%), 8 strains of Enterobacteriaceae(9.76%) and 5 strains of Corynebacterium(6.10%), respectively. The results proved that Staphylococcus aureus and Streptococcus agalactiae are the main pathogenic bacteria causing bovine subclinical mastitis in Eastern Hebei Province.

  • Prokaryotic Expression and Immunogenicity of Dominant Epitope Region of Goose Parvovirus Structural Protein VP3

    [Objective] The paper was to develop a subunit vaccine candidate for prevention and control of goose parvovirus infection. [Method]Based on the prokaryotic expression system, the antigenic epitopes and locations of the structural protein VP3 were predicted by software analysis,and the region displaying a large portion of antigenic epitopes was amplified by PCR. The target VP3 DNA fragment was inserted into pET-30 a-VP3 vector, was transformed into Escherichia coli BL21 competent cells for protein expression and animal test. The SPF chickens were immunized with the recombinant protein and the antisera were collected for neutralization test by using a goose embryo fibroblast. [Result] The recombinant plasmid was constructed, and the target region of VP3 protein was expressed efficiently in a soluble form. The neutralizing titers of antisera could reach up to-2.608. [Conclusion] The target region displaying a large portion of antigenic epitopes of the structural protein VP3 could be expressed efficiently in soluble form, and the expressed protein could induce neutralizing antibodies in SFP chicken.

  • Separation and Preliminary Identification of Intestinal Probiotics of Laying Hens

    [Objective] The paper was to isolate and identify probiotics in the intestine of laying hens. [Method] The intestinal probiotics in laying hens at peak period were isolated using conventional separation methods; the physical and chemical properties of target strains and in vitro antibacterial effects were measured. Moreover, the safety test of chicks was conducted. [Result] Four strains of lactobacillus were isolated from the jejunum of laying hens, including Lactobacillus plantarum, Bacillus acidophilus, L. delbrueckii subsp. Delbrueckii and L. delbrueckii subsp. Lactis, and their inhibition zone diameters were 18.30, 16.07, 11.45, 17.26 mm, respectively. One strain of Lactobacillus, L. brevis, was isolated from the cecum, with the inhibition zone diameter of 10.26 mm. Three strains of bacillus were isolated from the cecum, including Bacillus subtilis, B. cereus and B. licheniformis, and their inhibition zone diameters were 9.25, 8.46 and 8.37 mm, respectively. Daily drinking 2 billion units of viable bacteria was the safe dosage for chicks. [Conclusion] Eight strains of probiotics had certain inhibitory effect on Escherichia coli O_2, and had no toxic side effects to chicks. Lactobacillus had strong antibacterial effect on E. coli O_2, while the antibacterial effect of bacillus was relatively weak.

  • Influences of Lentinan on Production Performance and Some Immune Indexes of Broilers

    [Objective] The paper was to study the effects of lentinan on production performance and some immune indexes of broilers. [Method]Totally 200 1-day-old AA+ broilers were randomly divided into four groups, five repeats each group and 10 broilers each repeat. The broilers in control group, low dose group, middle dose group and high dose group were supplemented with the diet containing 0, 200, 350 and 500 mg/kg lentinan, respectively. The trial lasted 35 d. [Result] The average daily gains of broilers in low dose group, middle dose group and high dose group were significantly higher than that in control group at 14-21 days of age, 21-28 days of age and 28-35 days of age( P<0.05); the feed/gain ratios of broilers in low dose group, middle dose group and high dose group were significantly lower than that in control group at 21-28 days of age and 28-35 days of age(P<0.05); the immune organ indexes of broilers in low dose group, middle dose group and high dose group were significantly higher than that in control group at 35 days of age(P<0.05). [Conclusion] Adding lentinan in the diet could promote the production performance and immune organ index of broiler, and the addition amount of 350 mg/kg received the best effect.
