  • Preparation of Taenia solium Stained Specimens and Morphological Observation

    [Objective] In order to observe the morphological structure of Taenia solium and improve the dyeing method of Taenia solium. [Method]Taenia solium was stained by hematoxylin-eosin( HE) staining method and observed under microscope. Taenia solium specimens with clear structure and bright color were made. [Results] Male reproductive system and excretory system of Taenia solium segment could be observed under the optical microscope,structures of uterus and eggs in Taenia solium gravid proglottid could also be observed. [Conclusion]Aiming at providing a reference for the identification of Taenia solium and the diagnosis of the tapeworm disease.

  • Deposition Pattern of Inosine Monophosphate( IMP) in Pig Muscle during Cold Storage

    [Objective] The paper was to investigate the degradation and deposition changes of inosine monophosphate( IMP) and its metabolites in muscle of lean pigs and crossbreeding pigs under cold storage condition at 4℃. [Method]The contents of IMP and its metabolites in longissimus dorsi of lean pigs and crossbreeding pigs were determined by HPLC method. [Result]Duroc,Landrace,Yorkshire and DLY pigs shared the same degradation and deposition pattern on IMP,inosine and hypoxanthine. On the second day of cold storage,the content of IMP reached the maximum value,which increased by 0. 4%-15. 32% compared with the first day,and then it started to decline. On the fourth day of cold storage,the content of IMP significantly reduced by 23. 81%-39. 06% than that on the second day. On the fifth and sixth day of cold storage,the content of IMP kept on falling down slowly and maintained around 1. 0 mg/g. While the contents of inosine and hypoxanthine showed an increasing tendency with the extension of cold storage time. [Conclusion]Lean pigs Duroc,Landrace and Yorkshire and DLY threeway crossbreeding pigs shared the same degradation and deposition pattern on IMP and its metabolites. With the extension of cold storage,the content of IMP first increased then gradually decreased; while inosine and hypoxanthine gradually increased,and the difference among breeds was not significant.

  • Embryo Transfer by Surgical Method with Landrace as Receptor Sow

    [Objective] The paper was to test the feasibility of embryo transfer technique in pig production. [Method] Twenty-four estrus multiparity Landrace sows provided by Longjing Agricultural Science and Technology Institute were performed embryo transfer surgery,and postoperative effects were observed. [Result]Totally 11 out of 24 receptor sows were pregnant; pregnant sows delivered 67 cloned piglets,and the average farrowing rate of sows was approximately 6 piglets/sow.There were 22 mortalities of newborn piglets because of various reasons. [Conclusion] Embryo transfer technique is an indispensable link in pig production,and an important means of pig breeding and improvement.

  • Incubation Effect on Eggs of Tibetan Chicken under Low Altitude Condition

    [Objective] In order to study incubation conditions of Tibetan chickens in low altitude areas. [Method]Eggs of Tibetan chicken were transported to low altitude areas in highland areas for artificial hatching. [Results] The results showed that the average hatching rate of fertilized eggs was 85. 33%; the average healthy chick rate was 91. 77 %; the average primary body mass was 31. 21 g. Hatching rate of eggs in group 2 was the highest,which reached 90. 52%,the average egg mass was 43. 25 g and the average egg shape index was 1. 32. Hatching rate of eggs in group 2 was 13. 11% and 2. 47% higher than that in group 1 and group 3 respectively,which was 5. 19% higher than the average value. Hatching rate and healthy chick rate of eggs in group 2 also showed the same trend. [Conclusion] High hatching effect could be obtained under the better hatching condition in low altitude areas in highland areas.

  • Developmental Changes of GH Gene in Tan Sheep and Correlation Analysis with Slaughter Traits

    [Objective] The paper was to study the differential expression of growth hormone( GH) gene in different ages and different tissues of Tan sheep.[Method]Using real-time fluorescent quantitative PCR( RT-PCR) assay,specific primers were designed according to the GH gene sequence published on GenBank. With the total RNA of Tan sheep tissue as the template,the expression levels of GH gene in pectoral muscle,leg muscle and longissimus dorsi muscle at different ages were analyzed. [Result] In three types of muscle tissues,the expression patterns of GH gene in different tissues at the same month of age were pectoral muscle > leg muscle > longissimus dorsi muscle. The expression levels of GH gene were positively correlated with live weight before slaughter,carcass weight and net meat weight,and had significant positive correlation only with net meat weight( P < 0. 05). [Conclusion]The content of GH gene in different tissues gradually increased with the increasing months of age,and there were extremely significant differences in expression level of GH gene among different months of age or different sites( P < 0. 01).

  • Effect of Lactobacillus acidophilus 1.1878 on sIgA Content in the Intestinal Mucous and Antioxidant Capacity of Broiler Chicks

    [Objective] The paper was to study the immune activities of Lactobacillus acidophilus 1.1878 and its effect on antioxidant capacity of broiler chicks.[Methods]Samples of intestinal mucous,serum and liver were collected from broiler chicks after oral administration of L. acidophilus 1.1878. Secretory immunoglobulin A( sIgA) content in the intestinal mucous,total antioxidant capacity( T-AOC),level of MDA and activity of SOD and GSH-Px were determined,respectively. [Result] Oral administration of L. acidophilus 1.1878 significantly improved the s Ig A content in jejunum and ileum mucous( P < 0.05). No significant differences were observed in GSH-Px activity between treatment group and control group. The concentration of T-AOC and activity of SOD in serum and liver increased significantly( P < 0.05),while the level of MDA reduced significantly after oral administration of L. acidophilus 1.1878( P < 0.05). [Conclusion]L. acidophilus 1.1878 can enhance the immune function of intestinal mucous and antioxidant capacity of broiler chicks.

  • Preliminary Screening of Plant Ethanol Extracts Inhibiting IBV Proliferation

    [Objective] The paper was to study the inhibition effects of ethanol extracts from different plants on infectious bronchitis virus( IBV). [Method]Certain amount of plant ethanol extract was mixed with the recombinant virus IBV-3ab-luc inosculating luciferase gene at room temperature for 20 min,and then added in H1299 cell culture system together. The activity of luciferase was detected after 24 h,to compare the inhibition effects of ethanol extracts from different plants on the virus. [Result] The logarithms of Gynura segetum leaf,Prunella vulgaris powder,Plantago asiatica leaf,Ophiopogon japonicus root,Lycium barbarum fruit and Citrus reticulata were 4,3,3,2,0 and 0,respectively. [Conclusion]The ethanol extract from G. segetum leaf had the best inhibitory effect against IBV,followed by P. vulgaris powder,P. asiatica leaf,and O. japonicus root had relatively poor inhibition effect,whereas L. barbarum fruit and C. reticulata almost had no inhibition effect.

  • Observation on Culture Characteristics of Mycoplasma bovis with Four Different Antigen Quantification Methods

    [Objective] The paper was to explore the antigen harvest time of Mycoplasma bovis and the antigen quantification alternative method. [Method]M. bovis 08 M strain was inoculated in the Thiaucourt's medium containing 10% horse serum. Four growth curves were plotted by simultaneously measuring color change units( CCU),colony forming units( CFU),protein concentration and nucleic acid levels within 110 h. [Result]The growth of M. bovis was divided into four phases: the longarithmic phase appeared after being cultured for 10 h; the stationary phase appeared at 30 h with the highest number of viable cells up to1.0 × 108CCU/mL and 7.7 × 107CCU/mL,respectively; and the decline phase started at 75 h. The protein concentration of M. bovis increased rapidly from 15 to35 h,reached 72.06 μg/mL at 35 h,then maintained at 53.38-70. 65 μg/mL. The nucleic acid levels of M. bovis increased rapidly from 15 h,and the cycle threshold( Ct) values were maintained between 15.32 and 17.84 after 25 h. [Conclusion]There was a good correlation between the protein concentration and variable count of M. bovis at the early stationary phase,which was the best time period to harvest antigen. The protein concentration determination could be an alternative method to quantify antigen content of M. bovis when preparing inactivated M. bovis vaccine.

  • Enhancement of Humoral Immune Response of Newcastle Disease Vaccine by ATRA

    [Objective] The paper was to study the immune enhancement of ATRA on Newcastle Disease( ND) vaccine. [Method] The 1-day-old AA broilers were treated with ATRA at the doses of 1 and 5 μmol/kg,respectively. At 7 and 28 days of age,broilers in drug control group,low dose group and high dose group were immunized with ND vaccine by intranasal and eye immunization approach. At 7,14,21,28,35,42 and 49 days of age,seven chickens were randomly selected from each group and weighed. The thymus,spleen,bursa of fabricius and serum were collected for calculating immune organ index of thymus,spleen and bursa of fabricius. The ND specific antibody titers in serum were determined with HI test. [Result] ATRA promoted the growth of thymus,spleen and bursa of fabricius,and improved the immune organ index and ND specific antibody titers of chicks. [Conclusion]ATRA enhanced the humoral immune response of chicks,and ATRT at the dose of 5 μmol/kg presented more prominent immune enhancement effect on ND vaccine.

  • Study on PRRSV Receptor Genes Differential Expression in Lung Tissues in Different Breeds of Pigs after Infecting with HP-PRRSV

    [Objective] In order to study the susceptibility molecular mechanism of highly pathogenic porcine reproductive and respiratory syndrome virus( HPPRRSV) JXA1 isolate on Tibetan pig,Zangmei pig and Yorkshire pig. [Method]In the study,real-time quantitative RT-PCR method was established to compare and analyze the differential expression of five porcine reproductive and respiratory syndrome virus( PRRSV) receptor genes( HSPG2,SIGLEC1,CD163,VIM and NMMHC-II A) in lung tissues in Tibetan pig,Zangmei pig and Yorkshire pig before the challenge and at the 4~(th),7~(th) and 14 thdays after the challenge with JXA1 isolate. [Results] HSPG2 expression in Tibetan pig lung tissues increased significantly at the 4~(th) and 14 thdays after the challenge with JXA1( P < 0. 05),while decreased significantly at the 7~(th)day after the challenge( P < 0.05),HSPG2 expression in Zangmei pig lung tissues increased significantly at the 14 thday after the challenge( P < 0.05). SIGLEC1 expression in Tibetan pig lung tissues increased significantly at the 4~(th) and 14 thdays after the infection( P < 0.05),while SIGLEC1 expression in Yorkshire pig decreased significantly at the 4~(th),7~(th) and 14 thdays after the challenge( P < 0.05). CD163 expression in lung tissues of Tibetan pig and Zangmei pig both increased significantly at the 14 thday after the challenge( P < 0.05),while CD163 expression in lung tissues of Yorkshire pig decreased significantly at the 7~(th) and 14 thdays after the challenge( P < 0.05). VIM expression in lung tissues of Tibetan pig increased significantly at the 7~(th) day after the challenge( P < 0.05),while which of Yorkshire pig at the 7~(th) day after the challenge decreased significantly( P < 0.05). NMMHC-II A expression in lung tissues of Zangmei pig increased significantly at the 4~(th) day after the challenge( P < 0.05),and which of Yorkshire pig increased significantly at the 4~(th) and 14 thdays after the challenge( P < 0.05). [Conclusion]SIGLEC1 and VIM genes might be the potential key genes affecting the susceptibility of JXA1 isolate on Tibetan pig,Zangmei pig and Yorkshire pig.

  • Effects of Dietary Protein and Fat Levels on Growth Performance and Serum Biochemical Parameters of Male Mink

    This experiment was conducted to evaluate the effects of dietary protein and fat levels on growth performance and serum biochemical parameters of male minks during the growing period. A total of 90 healthy male minks at the age of 60 d were randomly assigned into six groups with 15 replicates per group and one mink per replicate. The minks were fed six experimental diets with two protein levels( 32% and 36%) and three fat levels( 10%,20% and 30%) for a 7-day preliminary period and then for 60-day experimental period. The results showed that the final body weight and average daily weight gain( ADWG) were significantly higher in low-protein,middle-fat group and low-protein,high-fat group than those in low-protein,low-fat group and high-protein,low-fat group( P < 0. 01). The final weight and ADWG were increased with dietary fat level increasing,and the two indices of 20% and 30% fat groups were significantly higher than those of 10%fat groups( P < 0. 01). The ratio of feed intake to weight gain( F/G) in low-protein,high-fat group was the lowest,which was significantly lower than that in lowprotein,low-fat group and high-protein,high-fat group( P < 0. 05). Serum urea nitrogen( UN) in low-protein,high-fat group was significantly lower than that in other groups( P < 0. 01). Serum alanine transaminase( ALT) activity in 32% protein groups was significantly higher than that in 36% protein groups( P < 0. 05). Serum triglyceride( TG) content in low-protein,high-fat group and high-protein,high-fat group was significantly higher than that in low-protein,low-fat group and high-protein,low-fat groups( P < 0. 05). Serum TG,total cholesterol( TCHO) and high-density lipoprotein( HDL) contents increased with the increase in dietary fat level,and they were all the highest in low-protein,high-fat group. The growth performance and serum biochemical parameters of minks were not affected by the interaction between dietary protein and fat levels( P > 0. 05). Considering all the factors,when the dietary protein is 32% and the fat level is20% to 30% under the condition of this experiment,the dietary protein and fat utilization rates are higher,and the male minks during the growing period can get a better growth performance.

  • Detection and Analysis of 1,2-propanediol Content in Whole-crop Silage Corn

    [Objective] The paper was to investigate the content of 1,2-propanediol in silage feed. [Method]Whole-crop corn was stored in laboratory bag silos and outdoor bunker silos. Bag silos were stored for 3,7,14,28,56 and 120 d,respectively,and fermentation products were analyzed after opening; the fermentation products,which were from three 500 t above-ground outdoor bunker silos stored for 120 d,were detected. Eight sampling points were selected for each bunker silo. [Result]No 1,2-propanediol was detected out in laboratory bag silos,and lactic acid was dominant in fermentation products. The acetic acid content significantly increased( P < 0. 05) with the extension of storage time,whereas the acetic acid content was kept below one-third of the lactic acid content; 1,2-propanediol and a small amount of 1-propanol were detected out in three bunker silos,and lactic acid was dominant in fermentation products,whereas the content of acetic acid was slightly lower than that of lactic acid. The differences in fermentation products from different silos and different sites in silos presented heterogeneity; except to propanediol,lactic acid bacteria and yeast,there was no significant difference in fermentation products at the top and bottom layers of silos( P > 0. 05); there were significant differences in contents of lactic acid,acetic acid,1,2-propanediol,and yeast between the outer and inner parts of silos( P < 0. 05); except to p H values and acetic acid,there were significant differences in fermentation products between bunker silos( P < 0. 05). [Conclusion]No 1,2-propanediol is detected out in laboratory bag silos due to relatively simple fermentation products. However,1,2-propanediol can be detected in the bunker silos due to complex outdoor environment for silage,and the microorganisms producing 1,2-propanediol exist in silos.

  • Effects of Yeast Chromium Combined with Probiotics on Laying Performance,Intestinal Microflora and Digestive Enzyme Activities of Laying Hens in Summer

    [Objective] To study the effects of yeast chromium combined with probiotics on laying performance,intestinal microflora and digestive enzyme activities of laying hens. [Method] A total of 432 healthy 40-week-old Roman brown laying hens were randomly divided into nine groups. Laying hens in control group were fed with basal diet,laying hens in test groups were fed with basal diet combining with different levels of yeast chromium( 0,400,600 μg/kg) or probiotics( 0,400,800 mg/kg). [Result]Yeast chromium,probiotics or yeast chromium combined with probiotics adding into feeds could significantly increased the laying rate( P < 0. 05) and significantly reduced feed-egg rate( P < 0. 05). Joint use of yeast chromium and probiotics had significant interaction effects on laying rate and feed conversion rate( P < 0. 05). Adding of yeast chromium combined with probiotics in diets could significantly increased the amount of Lactobacillus and Bifidobacterium( P < 0. 05) in ileum and cecum of laying hens,and significantly reduced the amount of Escherichia coli and Salmonella( P < 0. 05),in addition,the activity of total protease,lipase and amylase in duodenal contents were significantly increased( P < 0. 05). Joint use of yeast chromium and probiotics had significant interaction effects on the activity of total protease,lipase and amylase in laying hens duodenal contents( P < 0. 05). [Conclusion] Joint adding of600 μg/kg yeast chromium and 800 mg/kg probiotics in diets in summer had best effects on laying performance,intestinal microflora and digestive enzyme activities of laying hens.

  • Effect of Continuous Light on Rhythm of Apodemus agrarius

    [Objective] The paper aimed to study effects of continuous light on rhythm of Apodemus agrarius. [Method] The changes in rhythm of A. agrarius activities were observed under natural light and 24 h continuous light condition. [Result]A. agrarius activated at daytime and night,but the activity time at night was longer than that at daytime,and the peak period of activity also occurred at night. When exposed to natural light,the activity time,feeding time and drinking time of A. agrarius were( 27 870 ± 5 252) s,( 6 171 ± 1 529) s and( 474 ± 92) s,respectively; when exposed to 24 continuous light,the activity time,feeding time and drinking time of A. agrarius were( 19 935 ± 2 971) s,( 5 870 ± 1 988) s and( 331 ± 117) s,respectively. [Conclusion]The activity time of A. agrarius changed significantly under two different light conditions,while feeding time and drinking time had little changes.

  • Histology Observation on Intestine of Elaphurus davidianus

    Elaphurus davidianus is one of the rare species originally come from China. And study on intestine histology observation of Elaphurus davidianus has not been reported widely. In the test,microscopic morphology on intestinal tissue of Elaphurus davidianus was observed by the method of paraffin sectioning. Test results showed that jejunum epithelium villi structure of Elaphurus davidianus was obvious,a large number of intestinal glands distributed in lamina propria of jejunal epithelium. There was lymphoid tissue distributing in the base of the mucosa. The muscularis mucosa was composed of smooth muscle,thickness of muscularis mucosae was about 53 μm. There was no villous structure in the cecal mucosa of Elaphurus davidianus,capillaries in mucosal lamina propria were rich,the mucosal thickness was( 236 ± 55) μm. There were large amounts of intestinal glands in inherent layer,thickness of mucosal muscle was about 27 μm. The blood vessels located in submucosal loose connective tissue were rich. The structure of small intestine was similar with that of large intestine. There was no villus structure in rectal mucosal epithelium,the maximum thickness of mucosal layer was 833 μm,and the minimum thickness of mucosal layer was 313 μm. Mucosal muscular layer was developed,which was composed of several smooth muscle bands,different bundles scattered,the thickness was about 200-600 μm.

    In order to fully understand the distribution characteristics and geographical elements of fern flora and to reveal their process of natural history,the present study analyzed the floristic composition and geographical distribution of ferns in grassland of Gansu Province. The results showed that there were 31 species of ferns in grassland of Gansu,belonging to 19 genera and 14 families. The investigated ferns accounted for 22. 22%,8. 26% and 1. 99% of total families( 63),total genera( 230) and total species( 2 600) of ferns resources,respectively. The dominant families were Dryopteridceae and Polypodiaceae; the dominant genera were Selaginella and Woodsia. Families mainly belonged to tropical distribution type and genera mainly belonged to tropical and temperate distribution type without endemic genus. Vertical distribution was obvious; plant components contained old families and genera. The richness of fern in Gansu grassland was not high.

  • Effect of Laminarin on Growth and Immunity of Pearl Gentian Grouper

    [Objective] The paper was to study the effects of laminarin on growth performance and immunity of pearl gentian groupers. [Method] Four separate30 m2 concrete pools were used as a closed circulating water aquaculture system for the experiment. Simultaneously,approximately 1 800 groupers were placed in each of the two pools for treatment or control. The initial weight of the individual fish was( 275. 55 ± 29. 79) g. They were fed with either a control diet or one containing 0. 6% laminarin in the pools for 66 d. [Result]At the end of the feeding trial,the weight gain of the groupers in treatment group was increased by 29. 03%( P <0. 05),the specific growth ratio was increased by 23. 88%( P <0. 05),and the feed conversion rate was decreased by 15. 20%( P <0. 05) compared to control group,while there was no significant difference in muscle nutritional components between treatment group and control group( P > 0. 05). For each kilogram weight gain of an individual fish,the cost of the treatment diet was 1. 96 yuan less than that of control diet,i. e.,a reduction of 12. 54%. On the other hand,the activities of alkaline phosphatase( AKP),lysozyme( LZM) and total superoxide dismutase( T-SOD) of the groupers in treatment group were significantly higher than those in control group( P < 0. 05). [Conclusion] Addition of 0. 6% laminarin in the regular feed would improve the growth performance and immunity of pearl gentian groupers in aquaculture.
