  • A Little Thought for Pasture Informatization Construction

Compared with the countries with developed dairy industry worldwide,dairy industry in China is featured by not high informationization level and backward modernization level at the present stage. However,development and maturity of related technologies for pasture informatization construction provides a significant opportunity for transformation and upgrading and leaping development of dairy industry in China.

  • Genomic DNA Extraction Method for Mulberry Varieties in Anhui Province

    [Objective] The paper aimed to get a efficient,stable and low-priced DNA extraction method of mulberry leaves. [Method] With tender leaves of mulberry varieties in Anhui Province as materials,genomic DNA was extracted with CTAB method,and further identified by ultraviolet spectrophotometer,agarose gel electrophoresis and PCR methods. [Result] The A260/A280 ratio of extracted DNA was 1. 80-1. 90,and the yield was 0. 187-0. 275 μg/mg. With extracted DNA as the template,clear target bands were obtained by PCR. [Conclusion]Extracted DNA was good in quality,which could satisfy the needs of follow-up molecular biological operation.

  • Fattening Effects of Two Kinds of Three-way Crossbreeding Commercial Pigs

    To understand the fattening performance of Duroc × Landrace × Yorkshire and Duroc × Yorkshire × Landrace,48 individuals of two kinds of three-way crossbreeding lean type commercial pigs were selected to determine average daily feed intake,average daily gain and feed gain ratio. The backfat thickness and eye muscle area of pigs were measured at the end of trial. The results showed that Duroc × Landrace × Yorkshire and Duroc × Yorkshire × Landrace had similar fattening performance,and could be applied under scientific management conditions. The study is of great significance for development of pig industry.

  • Correlation Study on Body Size,Sexual Behavior and Semen Quality of Dezhou Donkey

    [Objective] The study was to explore whether the body size,sexual behavior of male donkey had certain correlation with reproductive traits. [Method] The data of body weight,body size and sexual behavior of male donkey during production were collected,combining with the semen quality data of ejaculation volume,density,motility and frozen-thawed semen motility were conducted the statistical analysis. [Results]( 1) Single ejaculation volume had a positive correlation with body height( r=0. 423,P=0. 044) and penis circumference( r=0. 495,P=0. 043),and total number of sperm per ejaculate had a positive correlation with body weight( r=0. 444,P=0. 036) and scrotal circumference( r=0. 520,P=0. 039);( 2) Ejaculation volume was negatively correlated with mount times( r=-0. 159,P=0. 035) and erection time( r =-0. 181,P=0. 023),semen density was negatively correlated with mating time( r=-0. 211,P=0. 004),and total sperm count was negatively correlated with erection time( r=-0. 159,P=0. 047). [Conclusion]The quality of semen had a certain correlation with body size,body weight and sexual behavior.

  • Comparison of pH Value, TBA Value, Glycogen and Lactic Acid Content in Muscle of Landrace and Yorkshire under Different Storage Time and Conditions

    In order to assess the meat quality under different storage time and conditions,the pH value,TBA value,glycogen and lactic acid content in muscle of Landrace and Yorkshire were measured. The results showed that within 10 h post slaughtering,post slaughtering time had extremely significant influence on pH value and lactic acid content of Landrace,but had no significant influence on glycogen content and TBA value. On the other hand,post slaughtering time had extremely significant influence on pH value of Yorkshire,but had no significant influence on glycogen,lactic acid content and TBA value. The pH values of Yorkshire at 3,5 and 10 h post slaughtering were 5. 21%( P < 0. 01),5. 64%( P < 0. 05) and 5. 59%( P < 0. 05) higher than those of Landrace,respectively; the lactic acid content in muscle of Landrace at 10 h post slaughtering was 56. 04%( P < 0. 05) higher than that of Yorkshire; the TBA value in muscle of Yorkshire at 10 h post slaughtering was 89. 19%( P < 0. 01) higher than that of Landrace; there was no significant difference in glycogen content between Yorkshire and Landrace. When pork stored at 4℃,storage time had no significant influence on pH value and glycogen content in muscle of Landrace,but had significant influence on drip loss,lactic acid content and TBA value; storage time had no significant influence on pH value,glycogen content and TBA value in muscle of Yorkshire,but extremely significant influence on drip loss and significant influence on lactic acid content. The pH value in muscle of Yorkshire at 24 h post storage at 4℃ was 6. 56%( P <0. 05) higher than that of Landrace; the glycogen contents in muscle of Landrace at 24,48,72,96,120 and 144 h post storage at 4℃ were 84. 90%( P <0. 05),78. 40%( P <0. 01),101. 87%( P <0. 05),83. 80%( P < 0. 05),83. 59%( P < 0. 05) and 67. 25%( P < 0. 01) higher than those of Yorkshire,respectively; the drip loss of Landrace at 72 h post storage and TBA value at 144 h post storage were 55. 70%( P < 0. 05) and 141. 33%( P < 0. 01) higher than those of Yorkshire,respectively; there was no significant difference in glycogen content between Yorkshire and Landrace. When pork stored at -20℃,storage time had significant influence on TBA value in muscle of Landrace,but had no significant influence on pH value,thawing water loss rate,glycogen and lactic acid content; storage time had no significant impact on pH value,thawing water loss rate,glycogen content and TBA value in muscle of Yorkshire,and had significant influence on lactic acid content. The pH value in muscle of Yorkshire at 24 h post storage at -20℃ was 5. 43%( P < 0. 05) higher than that of Landrace; the glycogen contents in muscle of Landrace at 24,48,72,96,120 h post storage at -20℃ were 85. 08%( P < 0. 05),108. 66%( P < 0. 01),72. 69%( P < 0. 05),90. 31%( P < 0. 01),70. 38%( P > 0. 05) higher than those of Yorkshire,respectively; the thawing water loss rates of Landrace at 24 and 72 h were 160. 14%( P <0. 05) and 74. 32%( P <0. 05) higher than those of Yorkshire,respectively; there was no significant difference in lactic acid content and TBA value at the same time point between Yorkshire and Landrace.

  • Effects of Compound Acidifier and Probiotics on Production Performance,Blood Biochemical Indexes,Antioxidant Indexes and Salmonella Infection of Laying Hens

    This study evaluated the effects of compound acidifier and probiotics on production performance,blood biochemical indexes,antioxidant indexes and Salmonella infection of laying hens. A total of 600 50-week-old healthy laying hens with similar laying rate were randomly assigned to four groups,10 repeats each group,and each repeat contained 15 chickens. The laying hens in control group were fed a basal diet,and in the other three experimental groups were fed basal diets supplemented with 300 g/t probiotics,0. 1% compound acidifier in the drinking water every other day,and 300 g/t probiotics + 0. 1% compound acidifier in the drinking water every other day,respectively. After the feeding of 16 weeks,a total of 60 laying hens from control group were divided into four treatments as the same above for a 13-day long Salmonella challenge( 1. 3 × 10~8 CFU) trial. The results were shown as follows: no significant effects were observed on production performance,hemoglobin content,the superoxide dismutase activity and total antioxidant capacity of reproductive organ in compound acidifier and probiotics groups( P >0. 05). Dirty egg rate was significantly decreased in compound acidifier group,probiotics group and combination group of both( P <0. 05). Compared with the control group,white blood cell count in probiotics group was significantly decreased( P < 0. 05),and the immunoglobulin content in probiotics group tended to be increased( P = 0. 061). The malondialdehyde( MDA) content in oviduct isthmus of hens in compound acidifier group and combination group was significantly decreased compared with the control group( P < 0. 05). The Salmonella positive rate in oviduct of hens in compound acidifier group was significantly decreased at the 7~(th) and 14~(th) days( P < 0. 05),but that in probiotics group and combination group was not significantly different compared with control group( P > 0. 05). In conclusion,compound acidifier and probiotics can improve health status of laying hens through decreasing the dirty egg rate and increasing antioxidant capacity and immunity of them,thus reducing Salmonella infection of body and eggs.

  • Expression Pattern and Target Gene of bbu-miR-103-1 in Buffalo(Bubalus bubalis) at Lactation and Non-lactation Periods

    [Objective] The paper aimed to investigate the expression pattern of bbu-miR-103-1 in buffalo( Bubalus bubalis) at lactation and non-lactation periods,and to predict its target gene and function. [Method]Expression pattern of bbu-miR-103-1 at lactation and non-lactation periods were detected by qRT-PCR.The precursor expression plasmid of bbu-miR-103-1 was constructed and named LpEZX-pre-miR-103-1. It was packaged and propagated to produce high-titer lentivirus in 293 T cell lines,which could be used to infect buffalo mammary epithelial cells( BMECs) and over express bbu-miR-103-1. The inhibitor of bbu-miR-103-1 was chemically synthesized and transfected into BMECs to suppress bbu-miR-103-1 at the same time. The relative expression of pantothenate kinase 3( PANK3) and milk fat metabolism related genes were detected by qRT-PCR. [Result]The relative expression of bbu-miR-103-1 at lactation period was 5. 29 times higher than that at non-lactation period in buffalo( P < 0. 01). The LpEZX-pre-miR-103-1 had been successfully constructed and packaged with the infection titer of 3. 47 × 10~6 PFU/m L. Overexpress or suppress of bbu-miR-103-1 extremely down-regulated or up-regulated the expression level of PANK3 in BMECs( P < 0. 01).Over expression of bbu-miR-103-1 extremely enhanced the expression of Acetyl-CoA carboxylase alpha( ACACA),Glycerol-3-phosphate acyltransferase 1 mitochondrial( GPAM),Diacylglycerol Oacyltransferase 1( DGAT1) and Pyruvate dehydrogenase lipoamide kinase isozyme 4( PDK4)( P < 0. 01),and also significantly up-regulated the expression of sterol regulatory element binding protein-1c( SREBP1c),Adipose differentiation-related protein( ADFP),Cluster of differentiation36( CD36),Acetyl-Co A synthetase short-chain subfamily member 1( ACSS1)( P < 0. 05). Over expression of bbu-miR-103-1 down-regulated the expression of PANK3,and improved the mRNA level of SREBP1c by feedback regulation,finally promoting the de novo synthesis of fatty acid beginning with ACACA.[Conclusion] bbu-miR-103-1 plays an important role in enhancing milk fatty acid synthesis,which provides a molecular base for revealing formation and regulatory mechanism of high-level milk fat in buffalo.

The result will provide technical support for preparation of scientific prevention and control measures against Chlamydia in Qinghai Province.

  • Survey of Porcine Viral Diarrhea in Beijing Using Umbilical Cord Blood

    [Objective] The study aimed to understand the status of piglet diarrhea virus in large-scale farms in surrounding areas of Beijing. [Method]By collecting 570 umbilical cord blood samples of diseased pigs,nucleic acids of porcine reproductive and respiratory syndrome virus( PRRSV),classical swine fever virus( CSFV),porcine pseudorabies virus( PRV),porcine circovirus( PCV),porcine epidemic diarrhea virus( PEDV),transmissible gastroenteritis virus( TGEV),and porcine rotavirus( RV) were detected by PCR or RT-PCR assay. [Result]The detection rates of PRRSV,PCV-2,PEDV,PPV,TGEV,PRV,RV and CSFV were 38. 7%,29. 6%,21. 8%,13. 03%,5. 6%,4. 2%,3. 8% and 1. 7%,respectively. [Conclusion] The major pathogens were PRRSV,PCV-2,PEDV and PPV,and mixed infection was serious.

  • Distribution of Heavy Metals in Common Energy Feedstuffs and Protein Feedstuffs in Sichuan Province

    [Objectives] The paper was designed to investigate the heavy metal distribution in energy feedstuffs and protein feedstuffs commonly used in Sichuan Province,in order to provide scientific basis for safe production of feedstuffs and animal products. [Methods] The contents of manganese( Mn),copper( Cu),zinc( Zn),vanadium( V),chromium( Cr),cobalt( Co),nickel( Ni),selenium( Se),molybdenum( Mo),cadmium( Cd),arsenic( As),mercury( Hg)and lead( Pb) in 68 samples of energy feedstuffs [corn,wheat,distillers dried grains with solubles( DDGS),corn bran with syrup,wheat bran and rice bran]and 61 samples of protein feedstuffs( corn gluten meal,corn germ meal,soybean meal,rapeseed meal and cottonseed meal) collected from Sichuan province were determined by inductively coupled plasma mass spectrometry( ICP-MS). [Results]( 1) The over standard rates of Cu,Cr and Pb in corn were 5. 00%,20. 00% and 5. 00%,respectively; and the means of over standard samples were 11. 00,1. 41 and 1. 06 mg/kg,respectively. The over standard rates of Cu,Zn,Cr,Se and Pb in wheat were 50. 00%,30. 00%,50. 00%,20. 00% and 30. 00%,respectively; and the means of over standard samples were 12. 96,54. 67,4. 92,0. 53 and 0. 72 mg/kg,respectively.( 2) The over standard rates of Cu,Zn,Cr,Se,Cd and Pb in wheat bran were 100. 00%,54. 55%,81. 82%,9. 09%,45. 45% and 72. 73%,respectively; and the means of over standard samples was 16. 01,62. 00,13. 96,0. 82,0. 42 and 1. 99 mg/kg,respectively. [Conclusions]The heavy metal pollution is more serious in grain by-products and cottonseed meal; the contents of Cr and Pb in feed ingredient are seriously excessive,which is potentially dangerous,and should be paid great attention.

  • Optimization on Production Key Factors of Microbial Fermented Feed and Analysis on Composition Variation During Fermentation

    [Objective] In order to study the production key factors of microbial fermented feed and the composition variation during fermentation. [Methods]Water,temperature,energy and protein were selected to study the effects on microbial fermented feed. And the fermentation conditions were optimized. Changes on nutrient composition during fermentation were determined and analyzed. [Result] Test results showed that feed formula with high sugar content,low protein,34%-36% water content,high fermentation temperature was more conducive to the production and actual production needs of fermentation strains. During the fermentation,strains showed interactions,test tended to be completed at the 144~(th)h after the fermentation,and number of lactic acid bacteria reached the peak of 1. 86 × 10~9 strain/g,contents of lactic acid could even reach 0. 89%. As time prolonged,contents of total energy,fiber and isothiocyanate gradually reduced,while contents of water and protein increased slightly. Contents of vitamin were stable,which in an order of V_B> V_E> V_K> V_A> V_D. Composition of amino acids showed an better change,contents of glutamic acid and proline decreased slowly,while contents of glycine and phenylalanine increased slowly. [Conclusion]High quality microbial fermentation feed could improve the palatability and safety of feed and keep the intestinal balance of livestock and poultry,which had broad application prospect.

  • Regulations of Antler Generation and Regeneration by Nutritional Factors

    Antler,one of the secondary sex characteristics of male deer,is a highly valued traditional Chinese medicine. The generation and regeneration of deer antler is regulated not only by hormones,but also by nutritional factors. Nutritional factors have great influences on pedicle generation,first antler generation and antler development. Therefore,this paper summarizes recent advances in the study of the influences of nutritional factors on antler generation and regeneration,analyzes the pathways regulating antler generation and regeneration by nutritional factors,and proposes that nutritional factors regulate antler regeneration by direct pathway and indirect pathway.
