  • Present Situation,Problems and Countermeasures for Harmless Treatment of Dead Livestock and Poultry in Fujian

    A serial of management and support policy for harmless treatment of dead livestock and poultry had been formulated in Fujian Province,and the effect was remarkable.But there were still some problems in the implementation of the policy,the technique of harmless treatment,livestock insurance and investigation and punishment of illegal behavior.Therefore,the following countermeasures were put forward: strengthen the responsibility system for harmless treatment of dead livestock and poultry; adjust harmless treatment mode to local conditions; build centralized treatment plant; punish illegal and criminal activities strictly;improve linkage policy of insurance and harmless treatment of livestock and poultry.

  • Residual Regularity in Tissues of Layer after Combined Exposure to Lead (Pb) and Cadmium (Cd)

    [Objective]The study was to investigate residual regularity in tissues of layer after combined exposure to lead (Pb)and/or cadmium (Cd).[Method]Lead acetate (100 mg/L)and/or cadmium chloride (50 mg/L)were administered in drinking water to 120 individuals of 40-week-old hyline layers for 8 weeks.[Result]Compared with the Cd group,the Cd content in various tissues,except kidney and femur,in Cd+Pb group all increased,but the difference was not significant (P>0.05).The Cd contents in kidney and femur in Cd group were significantly higher than that in Cd+Pb group.Compared with the group Pb,the Pb contents in breast muscle and kidney in Cd+Pb group decreased significantly (P<0.05);the Pb contents in femur decreased extremely (P<0.01);while the Pb contents in leg muscle,heart and spleen increased significantly (P<0.05);the Pb contents in liver and glandular stomach were almost the same.Compared with the control group,the Cd content in various tissues in Pb group increased significantly (P<0.05);no significant changes were observed for Pb content in Cd group.[Conclusion]Deposit of Pb and Cd in poultry body had complex interaction.

  • Differential Expression of CCNA2 Gene and MMP2 Gene in Skin Tissue of Rex Rabbit with Different Fur Densities

    [Objective] The paper was to search for the molecular marker highly associated with fur density of rex rabbit.[Method] Experimental rex rabbits were divided into dense fur group and sparse fur group.The variation rule of gene expression in skin tissue of rex rabbits with different fur densities was detected using rabbit whole gene c DNA microarray.[Result] A total of 2 657 genes with differential expression were detected by microarray test,including 1 106 genes with known function,687 genes with up-regulated expression and 419 genes with down-regulated expression.The expression levels of multiple genes involving in cell proliferation,differentiation,apoptosis and migration had significant changes.MMP2 gene had high expression in skin tissue of rex rabbits in dense fur group,which was 8.195 times of gene expression in sparse fur group; CCNA2 gene had low expression in skin tissue of rex rabbit in dense fur group,which was0.143 times of gene expression in sparse fur group.[Conclusion] The results provide the theoretical basis and research direction for early breeding of rex rabbit in practical production.

  • Complete Sequence Determination and Origin of Mitochondrial DNA Control Region in Dehua Black Chicken

    Primers were designed according to the genomic DNA of red jungle fowl,and the complete sequence of mt DNA D-loop region in Dehua black chicken was obtained.Combined with the D-loop region complete sequence of red jungle fowl in Gen Bank,the maternal origin of Dehua black chicken was explored.The results showed that the complete sequence of mt DNA D-loop region in Dehua black chicken was 1 231 bp,and the average contents of T,C,A and G were33.5%,26.5%,26.7% and 13.3%,respectively.Phylogenetic analysis showed that Dehua black chicken clustered with red jungle fowl (Gallus gallus spadiceus),suggesting Dehua black chicken probably originated from G.g.spadiceus in Yunnan Province or nearby areas.

  • Polymorphism of POMC Gene in Guizhou Yellow Cattle

    [Objective]The paper was to explore the effect of pro-opiomelanocortin (POMC) on feeding behavior and energy homeostasis of animals.[Method]Using DNA sequencing technology,polymorphisms of POMC gene in 92 individuals of Guizhou yellow cattle were studied.[Result]Five mutation sites,138 C> T,430 C> T,644 C> T,668 C> T,693 A> G,were found in the 870 bp amplification product.Polymorphism results showed that except the mutation site 430 C>T that was deviated from the Hardy-Weinberg equilibrium (HWE) state (P<0.05),the rest mutation sites were at the HWE state (P>0.05); the peptide information content (PIC) of five mutation sites were moderate polymorphism.[Conclusion]The POMC gene of Guizhou Yellow Cattle was abundant in genetic diversity,with great selection potential,which could be used as a potential genetic marker of cattle growth traits.

  • Expression of ChIFN-α in Pichia pastoris and Optimization of Its Expression Condition

    Because yeast codon preferred to synthesize the gene sequence of Ch IFN-α mature peptide,Pichia pastoris expression system was connected into vector p PIC9K,to construct recombinant plasmid p PIC9K-α.After digestion and linearization,the recombinant vector was cloned into P.pastoris GS115,high copy transformant and its methanol utilization type were screened,and its expression conditions were optimized.The results showed that Ch IFN-α successfully expressed in P.pastoris,the expression amount of recombinant protein was the largest after induced expression in 0.5% methanol containing 2% acid hydrolyzed casein at 28℃ for 72 h,and its antiviral activity was the highest.

  • Epidemiological Investigation of Avian Leukemia in Some Indigenous Chicken Breeds of China

    [Objective] The paper was to understand the natural infection status of avian leukemia in some indigenous chicken breeds of China.[Method]Using ELISA assay and virus isolation method,epidemiological investigation of ALV-AB and ALV-J avian leukemia of 10 indigenous chicken breeds were conducted.ALV dynamics were monitored in F_2 generation of four chicken lines.ALV pollution of attenuated live vaccines used in raising process was also inspected through sampling method.[Result] The positive rate of ALV-P27 antigen was 0-62.1%; the positive rate of ALV-AB antibody was 0-25.0%; the positive rate of ALV-J antibody was 0-59.0%; the positive rate of virus isolation was 0-22.0%.The positive rate of ALV-P27 antigen in 1-day-old chick meconium of four lines was 6.0%- 67.0%.The positive rate of virus isolation in 6-week-old chickens was 2.0%- 34.3%.Two kinds of vaccines from two batches produced by a manufacturer were polluted by ALV.[Conclusion] Most indigenous chickens were infected by ALV.There were great differences among different breeds of indigenous chicken,which might be related to ALV genetic resistance of different indigenous chickens.The ALV positive rates of F_2 chicks were slightly enhanced in some lines,which might be related to vaccine pollution.

  • Effects of Different Antibiotics Substitutes on Production Performance of Fattening Pigs

    [Objective] The aim was to discuss effects of different antibiotics substitutes on production performance of fattening pigs,and to lay a foundation for development and application of antibiotics-free pig feed.[Method]By adding 100 mg/kg compound probiotics,100 mg/kg compound enzyme preparation,500 and 1 000 mg/kg eu-commin in the basal diet,binary hybrid pigs weighing about 15 kg were reared for 102 d,and the feed added with 300 mg/kg colistin sulfate was used as control.[Result] Using compound probiotics and compound enzyme preparation to substitute antibiotics had no adverse effects on daily weight gain,daily feed intake and feed gain ratio of 15-90 kg fattening pigs; using eu-commin to substitute antibiotics had no adverse effect on daily weight gain,daily feed intake and feed gain ratio of 15-30 kg fattening pigs,but had significant influence on daily weight gain of 30- 90 kg fattening pigs (P<0.05).[Conclusion] It is feasible to develop antibiotics-free pig feed by using new type of green environmental-friendly additive instead of antibiotics.

  • Preventive Effects of Five Drugs on Mycoplasma Pneumonia of Swine (MPS)

    The paper was to explore the preventive effects of five drugs on mycoplasma pneumonia of swine (MPS) and to provide reference for clinical medication of pig farms in Hainan Province.A total of 444 health piglets were randomly divided into 6 groups,including five medication groups (72 piglets in group A,74 piglets in group B,72 piglets in group C,76 piglets in group D,76 piglets in group E) and one control group (74 piglets).The piglets in experimental groups were treated drugs once a day for successive 5 days at 30,60,90,120 and 150 of age.The piglets in control group were free of medication.At 70 and 140 days of age,15 piglets of each group were randomly selected to collect their blood serum.The Mycoplasma hyopneumoniae (M-Hyo) antibodies in serum were measured by enzyme-linked immunosorbent assay (ELISA).During the experiment,the incidence rates of respiratory disease,lung lesion,feed conversion rate,average daily gain (ADG),and mortality rate of pigs were also observed and recorded.The results showed that the five drugs had significant difference in preventative effects.Group C (Zhiyuanjing group) received the best preventive effect and the highest economic benefits.Compared with control group,the ADG and feed conversion rate in group C were increased by 7.53% and 9.09%,respectively; the incidence rate of respiratory disease was reduced by 13.44% and lung lesion was alleviated by81.43%; and the earnings of each pig could rise by 132.70 yuan.The preventative effect and economic benefit of the drugs was sequenced by Chansu Kechuanling and Bingchan Kechuanwang.Wante Feilin and amoxicillin had weaker preventive effects against MPS but greatly influenced growth performance of pigs,so they should be used alternatively with other drugs.

  • Effects of Shuanghuanglian Propolis Oral Liquid on Growth Performance and Intestinal Microflora of Broilers

    [Objective] The paper was to investigate the effects of adding different levels of Shuanghuanglian propolis oral liquid in drinking water on growth performance and intestinal microflora of broilers.[Method] Ninety one-day-old broilers were randomly divided into three groups (group I,group II and group III) : group I was the control group,and groups II and III were added with 0.5 and 1.0 mL/L Shuanghuanglian propolis oral liquid in drinking water,respectively.[Result] Compared with broilers in group I,the average daily gains of broilers in group II and group III increased significantly (P<0.05); the feed gain ratio of broilers in group III was significantly lower than that in group I (P<0.05); the number of intestinal Lactobacillus spp.in group II and group III increased significantly (P<0.05); the number of Salmonella spp.and Escherichia coli in group III decreased significantly (P<0.05); the number of Bifidobacterium spp.increased significantly (P<0.05).[Conclusion]Adding 0.5 and 1.0 mL/L Shuanghuanglian propolis oral liquid in drinking water could improve growth performance and intestinal microflora of broilers,and the dose of 1.0 mL/L is recommended in clinical application.

  • Effects of Seeding Rate,Water and Fertilizer Coupling on Grass Yield of Forage Millet (Setaria itlica) in Hebei

    The paper was to study the effects of seeding rate,water and fertilizer (N,P,K) coupling on grass yield of forage millet Jigu No.18 (Setaria itlica).A quadratic regression orthogonal rotation combination with five factors was designed in pot experiment.The mathematical model between hay yield of forage millet (Y) and soil moisture content (x_1),N fertilizer (x_2),P fertilizer (x_3),K fertilizer (x_4) and seeding rate (x_5) was established to simulate optimization.The results showed that moisture content,seeding rate,P fertilizer and K fertilizer had important effects on hay yield.Soil moisture content had the biggest impact on yield,followed by seeding rate,P and K fertilizer.The coupling effects of various factors successively were moisture content/seeding rate > K fertilizer/seeding rate > N/P fertilizer > soil moisture/N fertilizer > soil moisture/P fertilizer.Moreover,the mathematical model,Y=20 543.756-565.570x_1- 39.942x_2- 23.102x_3- 38.470x_4- 151.877x_5+ 1.052x_1x_2+ 1.604x_1x_3+ 12.953x_1x_5- 0.173x_2x_3+ 0.737x_4x_5- 2.292x~2_5,was established.The optimum soil moisture and seeding rate were determined as 10% and15 kg/hm~2,respectively.In this scheme,the hay yield was 14 037.151 0 kg/hm~2 and the economic benefit was 13 887.15 yuan/hm~2; the income was increased by 23.68% (3 288.98 yuan/hm~2) compared to the optimal combination in the test.The results provided a theoretical basis and technical support for forage millet production in Hebei Province.

  • Breeding and Application Prospects of a New Quinoa Variety Longli 1

    Longli 1 (Chenopodium quinoa Willd) was the first new quinoa variety in China,which was bred by Pasture and Green Agriculture Institute of Gansu Academy of Agricultural Sciences through the methods of line breeding,cultivation and domestication.In multipoint regional test,the average yield of Longli 1 was2 100.00 kg/hm~2,which was increased by 9.6% compared to Jingle quinoa.Longli 1 was a mid-late mature quinoa variety.The plant was broom-shape,with the plant height of 181.2- 223.6 cm,the growth period of 128- 140 d,and the branch number of 23-27. The seeds were round pill shaped,with 1 000-grain weight of 2.40- 3.46g.The contents of crude protein,fat,proline and total phosphorus in seeds were 171.5- 187.8,56.5- 59.3,5.5- 6.9 and 4.5- 6.8g/kg,respectively.Longli 1 presented resistance against downy mildew and leaf spot,with strong disease-resistant ability.In multipoint regional test and production test,Longli 1 displayed lodging resistance with strong regeneration ability.It also presented resistance to cold,drought,saline and alkaline,and barren,with wide adaptability.Longli 1 was suitable for planting in mountains,plain field and irrigation area with frost-free period greater than 120 d,rainfall more than 250 mm,and altitude 1 500- 3 000 m.The application prospects of quinoa were also put forward in the paper.

  • Production Performance of Different Varieties of Oat (Avena sativa) in Winter Dry Land of Hunan

    The phenological phase,plant height,tiller number,nutritional composition,yield and stem/leaf ratio of three oat (Avena sativa) varieties (lines)in winter dry land of Hunan were studied.The result shows that the three oat varieties were greatly different in plant height,nutritional composition,yield and stem/leaf ratio.Baiyan 7 had the best performance,with the plant height,tiller number and fresh yield of 101.67 cm,7.6 tiller/plant and 70.15 t/hm~2 respectively; the crude protein content of dry matter of Baiyan 7 was 32.63%; the leaf weight per tiller of Baiyan 7 accounted for 31.6% of single tiller weight,while those of Baiyan 8 and Baiyan 2 accounted for 31.3% and 29.2%,respectively.The single tiller weight of oat could be estimated/calculated by the model/formula Y=0.043 7X-2.89 (R~2=0.913 4,P<0.01),where Y is the single tiller weight (g) and X is the plant height (cm).Comprehensive analysis showed that Baiyan 7 had higher yields and stem/leaf ratio.Thus,Baiyan 7 is more suitable for planting as a winter-spring forage in the winter dry land of Hunan.

  • Study on Growth,Serum Biochemical and Antioxidant Enzyme Indices of Juvenile Siganus guttatus Fed with Enteromorpha prolifra and Artificial Feed

    In order to provide reference for using Siganus guttatus to control overgrowth of Enteromorpha prolifra,the growth,serum biochemical and antioxidant enzyme indices of juvenile S.guttatus respectively fed with E.prolifra and artificial feed were studied.One hundred and eighty individuals were cultivated for 90 days in six 2.5 m × 1.5 m × 1.5 m cages (30 ind.per cage) which mesh size were 0.5 mm.The experimental animals were divided into two dietary groups (three cages for each group) that were fed with E.prolifra and artificial feed respectively.During the culture period,temperatures ranged from 23.0 to 26.5℃,p H was between 7.8 and 8.2,dissolved oxygen was more than 5.0 mg/L.The weight gain rate,specific growth rate,relative growth rate,hepato-somatic index of juvenile S.guttatus fed with E.prolifra were significantly lowe than that fed with artificial feed (P<0.05).The total protein (TP),urea nitrogen (UN)and alkaline phosphatase (AKP) of juvenile S.guttatus fed with E.Prolifra were significantly higher than that fed with artificial feed (P<0.05),while glutamic oxalacetic transaminase (AST/GOT) and glutamic-pyruvic transaminase (ALT/GPT) of juvenile S.guttatus fed with E.prolifra were significantly lower than that fed with artificial feed (P<0.05).SOD,GPX and anti-superoxide anion radical activity in liver,kidney,gill and muscle of juvenile S.guttatus fed with E.prolifra were higher than that fed with artificial feed,and these antioxidant enzyme activities in kidney of juvenile S.guttatus fed with E.prolifra was significantly higher than that fed with artificial feed (P<0.05),while CAT and hydroxyl radical-inhibiting activity in liver of juvenile S.guttatus fed with artificial feed were significantly higher than that fed with E.prolifra (P<0.05).Juvenile S.guttatus fed with E.prolifra showed poor growth performance but better antioxidant defense system.S.guttatus is easily raised and has stronger ability of digestion and absorption of E.Prolifra.The technique that uses S.guttatus to control E.prolifra deserves deeply study.

  • Molecular Cloning and Bioinformatics Analysis of kdpE Gene from Vibrio alginolyticus

    The two-component regulatory system plays an important role in the growth and virulence of bacteria.In this study,the phosphate transcriptional regulatory protein (KdpE) gene of Vibrio alginolyticus strain HY9901 was cloned.Sequence analysis revealed that the length of kdp E gene is 732 bp and encodes a putative protein of 243 amino acids.The predicted molecular weight (MW) of Kdp E was 27.66 k D with an estimated p I of 5.01.Using SignalP 4.0 and TMHMM Server 2.0 software,it was predicted that the Kdp E protein was located in cytoplasm.It did not contain a signal peptide or a transmembranous region.A phylogenetic tree was constructed by MEGA 5.0 software,indicating that the Kdp E of V.alginolyticus had high genetic relationship with Vibrio campbellii and Vibrio parahaemolyticus.Using SWISS-MODEL work-space,the three-dimensional structures of conserved domain REC in the Kdp E was determined.These results can provide a basis for further studies on the two-component regulatory systems of V.alginolyticus.

    In recent years,artificial rearing snake diseases are increasingly rising,and some farms have suffered huge losses due to outbreak of snake diseases.Disease has become a big bottleneck restricting artificial breeding industry of snakes,and effective prevention and control of snake diseases has become the top priority.To control snake diseases purposely,questionnaires of artificial rearing snake diseases were released to 21 snake households in Yongzhou area of Hunan Province from February to May in 2014,and on-the-spot investigation was conducted as well.The types,etiology,epidemic characteristics,clinical symptoms and control effects of snake diseases in Yongzhou area were preliminarily understood,and their causes were analyzed.Survey showed that dermatitis,pneumonia,stomatitis,parasitosis,enteritis and powdery mildew were the six major diseases of artificial rearing snakes in Yongzhou area.The incidence of dermatitis was the highest of 61.9%,followed by pneumonia (42.8%).
