[Objective]The aim was to to establish a kind of peculiar, sensitive and quick fluorescent PCR detection method. [Method]A peculiar, sensitive and quick method of fluorescent PCR detection for avian infectious bronchitis virus was established, the standard curve was built, specific primers, susceptibility and repeatability was detected.[Result]This method diagnosed avian infectious bronchitis virus peculiarly, sensitively and quickly, simple and easy to use, time short, suitable for clinical testing. [Conclusion]This research laid the foundation to diagnose avian infectious bronchitis virus.

[Objective] This paper aimed to sum up the clinical symptoms, pathological changes, diagnosis and control method of swine streptococcicosis. [Method] Ill and dead pigs were dissected and organs such as liver, kidney, spleen and lung were aseptically took to inoculate in sheep blood agar, respectively, with bacteria isolated and purified to Gram staining and microscopy. [Result] It was positive in Gram staining and hemolysis ting was formed on the sheep blood agar; the bacteria could ferment sucrose, sorbital, glucose, mannose, lactose and hydrolyze sodium hippurate. [Conclusion] Through drug sensitive test, it indicated that swine streptococcus suis was highly sensitive to cephalosporin, ciprofloxacin and penicillin, and showed resistance to kanamycin, oxytetracycline, streptomycin and gentamicin.

[Objective] This experiment aimed to find out the origin and genetic evolution relationship of chicken flavivirus (CFV) CJD05 strain in Fujian Province. [Method] A pair of primers were designed and synthesized according to the sequences of E gene from Duck flavivirus (DFV) isolate BYD-1. E gene of CFV isolate CJD05 was specially amplified and its sequences were analyzed. [Result] The target bar which was cloned from CFV isolate CJD05 was 1 503 bp. Homology analysis was conducted to compare E gene nucleotide sequence of CFV isolate CJD05 with DFV isolate BYD-1 and goose flavivirus (GFV) isolate JS804. Results indicated that isolate nucleotide homologies were 99.2% and 99.3%, and amino acid homologies were 99.0% and 98.6% respectively. [Conclusion] CFV isolate CJD05, DFV isolate BYD-1 and GFV isolate JS804 were highly homologous. The homology of CFV isolate CJD05 with Tembusu virus (TMUV) was higher than with other arthropod-borne flaviviruses.

[Objective] The paper aimed to provide more information for effective prevention and control of clinical canine diarrhea. [Method] 156 cases of dogs with clinical diarrhea were selected as objects to analyze the reasons for its diarrhea and observe the therapeutic effect. [Result] Bacterial and viral diarrhea are major causes of canine diarrhea; The cure rate of parasitic diarrhea was high while that of viral diarrhea was poor; That probiotics and white clay combined with conventional therapies had a good therapeutic effect on canine diarrhea. [Conclusion] It was essential to keep exploring more effective therapies to treat canine diarrhea matched with basic therapies.

[Objective] To establish an indirect ELISA method which can detect fowl adenovirus group I (FAVI) antibody easily and rapidly. [Method] The expressed and purified FAVI penton recombinant protein was used to be an antigen, optimized the reaction conditions, and then established the FAVI indirect penton-ELISA antibody detection method. [Result] The optimal coating concentration of antigen was 1.5 μg/hole, the optimal coating condition was 37 ℃ 2 h and 4 ℃ overnight; the optimal dilution of serum was 1∶ 100; the optimal working concentration of anti-chicken IgG-HRP was 1∶2 000; the positive and negative critical value of ELISA was 0.335. Detected the 100 chicken serum samples by the established penton-ELISA method, the positive rate was 41%. [Conclusion] Through the study, the established penton-ELISA method has a good specificity, sensitivity and reproducibility. And it offers an effective tool for the diagnosis of FAVI, the survey of antibody and epidemiology survey.

[Objective] The research aimed to discuss the relationship between the polymorphism of PPARγ-2 gene and the susceptibility of type 2 diabetes mellitus (T2DM) in Guangxi Bama mini-pigs. [Method] 24 Guangxi Bama mini-pigs were fed with high-fat and high-sucrose diet, and partial sequences of exon 2 of PPARγ-2 gene were amplified by using PCR method. In addition, the contents of fasting blood glucose and insulin (INS) in Guangxi Bama mini-pigs were determined, and the glucose tolerance test (GTT) was also carried out. [Result] There was one SNP site (19813A/G) in partial sequence of exon 2 of the cloned PPARγ-2 gene, and AA (7 pigs) and AG (17 pigs) genotype were detected. The contents of fasting insulin and 60-min blood glucose in GTT in AG-genotype Guangxi Bama mini-pigs were significantly higher than those of AA genotype (P<0.05), while the incidence of T2DM in AG-genotype Guangxi Bama mini-pigs (71.4%) was obviously higher than that of AA genotype (5.9%). [Conclusion] The polymorphism of 19813A/G in exon 2 of PPARγ-2 gene was related with the susceptibility of T2DM in Guangxi Bama mini-pigs.

[Objective] To improve the accuracy and efficiency of the detection which used in Perkinsus sp and Marteilia refringens, and then shorten the detective cycle.[Method]According to the gene sequence of Perkinsus sp and Marteilia refringens from gene bank, design two pairs of specific primers and two TaqMan probes with different fluorophores labeled. Optimizing the reactive conditions and reagent concentration in order that establishing the duplex real-time PCR method for detecting Perkinsus sp and Marteilia refringens simultaneously.[Result] The sensitivity of the duplex real-time PCR method which about Perkinsus sp and Marteilia refringens is 40 template copies. After combine the templates of Perkinsus sp and Marteilia refringens with different concentrations, this method still could be detect this two protozoan efficiently and synchronously.[Conclusion] The established duplex real-time PCR method for detecting Perkinsus sp. and Marteilia refringens possesses lots of advantages, such as specific, sensitive, rapid, quantitative and reproducible, can be used for clinical detection of infection which was caused by Perkinsus sp. and Marteilia refringens.

[Objective] The aim of this study was to evaluate the effect of livestock exclusion on the health of grassland ecosystem accurately and quantitatively. [Method] One open grazing site and five seasonal exclusion sites, which had the same vegetation composition (taking Leymus chinensis and Stipa krylovii as main species) and soil type, were chosen as research objects for the VOR and CVOR evaluation of the effect of open grazing and livestock exclusion on the health of typical steppes, and for building the calculation model for VOR and CVOR indices to evaluate the health of typical steppes with different exclusion periods. [Result] The VOR and CVOR composite indices both could accurately evaluate the health of typical steppes, and the evaluation results from the both indices were quite consistent. Although the condition(C), vigor(V), organization(O) and resilience(R )indices changed differently in different exclusion periods, the VOR and CVOR composite indices increased gradually with the period of exclusion prolonged, and the health condition of typical steppe changed from "alert" level (in the open grazing and earlier exclusion period) to "healthy" level (after 13 to 20 years' exclusion). [Conclusion] The application of exclusion was beneficial to the recovery of degraded grassland ecosystem.

[Objective] The experiment aimed to check the pigs' digestive utilization of phosphor and other nutrient components in fermented soybean meal. [Method] 15 8-week old two-way cross growing pigs (average weight was 23.6 kg) were selected and divided randomly into 3 groups, 5 pigs per group. The method of total feces collection was adopted to compare the digestibility of phosphor and other nutrient components in growing pigs fed respectively by fermented soybean meal by Aspergillus usami and normal soybean meal. The experimental design were normal soybean group (NS group, P: 0.087%), fermented soybean meal group (FS group, P: 0.089%), normal soybean+ fermented soybean meal mixing group (MS group, P: 0.089%). [Result] The results indicated that the digestibility of growing pigs in FS group was significantly higher than that of NS group. Meanwhile, the digestibility of crude protein was significantly improved. [Conclusion] The above results indicated that Feeding with soybean meal fermented by Aspergillus usami not only could increase the digestibility of phosphor and protein, but also decrease the excretion of phosphor and nitrogen.

[Objective] To study the effect of feed intake characteristic on growth performance in Luyu-Duroc and Yorkshire. [Method] 89 Luyu-Yorkshire and 82 Luyu-Duroc were chosed and fed in the automatic determination system for swine production, 24h real-time monitoring feed intake traits and growth performance. [Result] It showed that: as feed intake increased, average daily gain (ADG) and weight of 4 months were enhanced, days on test, feed conversion ratio (FCR) and age-corrected of weight at 100 kg were reduced, feed intake positively regulated growth performance of Luyu-Doroc and Yorkshire. Feed intake significantly impacted ADG and days on test (P<0.01), notably affected age-corrected of weight at 100 kg of Luyu-Duroc and FCR of Luyu-Yorkshire (P<0.05), variation tendency of feed frequency was consistent with feed intake and feed time, and the correlation was significant. [Conclusion] The correlation between feed intake traits and growth performance was mainly caused by feed intake, ADG and FCR, however, predictability of feed intake was stronger than ADG and FCR, so in the actual selection feed intake was increased in pig breeding, thus at the same time to enhance ADG and to reduce FCR.

[Objective] To study the antioxidant effects of LC on D-galactose induced aging mice. [Method] 50 mice were randomly divided into blank control group, D-galactose model group, LC treatment groups at three levels of 0.25, 0.5 and 1.0 g/(kg·d), a total of 5 groups. Using the subacute D-galactose induced aging models, the mice were killed after 6 weeks of experiment and the activity of Superoxide dismutase (SOD), Glutathione peroxidase (GSH-Px) and Malondialdehyde (MDA) content in plasma, brain and liver tissue were determined. [Result] The activity of SOD and GSH-Px in plasma, brain and liver tissue were significantly increased after treated with LC, while the content of MDA were decreased with a dose-dependent manner. [Conclusion] LC had anti-aging effects in mice, and the mechanism may be correlated with the antioxidation.

[Objective] This paper aimed to summarize the results of the lenok reproduction in a hatchery, including growth of the artificially bred lenok, artificial propagation and fry rearing. [Method] Lenok (Brachymystax lenok) collected at wild were fed with pelleted feed and cultured to maturity in a flowing-water pond. The broodstocks were hormonally induced to spawn, and fry were reared from 2003 to 2010 in order to obtain a suitable and reliable method for the mass production of lenok juveniles. The embryos were hatched at accumulative temperature of up to 180-213 day degree at water temperature of 8-10 ℃. [Result]The results showed that 68%-85% of success rate was obtained with the eyed rate (fertilizing rate) of 32.8%-83%, hatching rate of 56.5%-87%, and emerging rate of 59.9%-90%. The fry developed into swim-up stage 10-12 days post-hatching. There were no significant differences in growth rate, fecundity and the eyed rate in the cultured lenok from those of the wild lenok. [Conclusion] Mass fry production of lenok could be achieved by hormonally induction.

[Objective] The aim of this study was to determine the effects of dietary phospholipids on Na+-K+-ATPase activity and cell membrane fatty acid composition in the gill of catfish (Pangasius sutchi). [Method] 900 healthy catfish (1.4±0.08 g of average weight) were randomly divided into five groups. Dietary phospholipids level of group PL0 was 0%, and 1%, 2%, 3%, 4% for group PL1, PL2, PL3, PL4, respectively. The experiment lasted for 56 days. [Result] The results showed that the Na+-K+-ATPase activity in gill decreased gradually with the increase of the dietary phospholipids level (P<0.05). Cell membrane fatty acid composition was affected significantly by dietary phospholipids. The content of saturated fatty acids and monounsaturated fatty acids of group PL0 was the highest (P<0.05), however, it had the lowest content of highlyunsaturated fatty acids (P<0.05). The content of polyunsaturated fatty acids of group 3 was higher than group 0 and group 1 (P<0.05). [Conclusion] The results indicated that there is obvious compensation effects on Na+-K+-ATPase activity with phospholipids content changes in gills of catfish (Pangasius sutchi) to maintain stable physiological level and normal basal metabolism and the increase of ΣHUFA and ΣPUFA is necessary for gill cell membrane to better maintain and perform normal physiological function.
