• Migration Time and Distribution of Porcine Embryos in Maternal Uterus

    [Objective] The aim of this study was to determine migration time of porcine embryos and to investigate the effects of embryo number on migration and spacing of porcine embryos. [Method] Migration and spacing of embryos were observed in gilts with one occluded oviduct and in those with one ovary. Their uteri were observed on Day 6, 7, 8, 9, 10, 11, 12, 13 and 15 of gestation, respectively. [Result] The embryos migrated from oviduct at one side to uterine horn at the other side. On Day 8 and 9, the embryos first migrated from one horn to the other horn. On Day 15, they could occupy the uterus completely. Migration rate and distribution of embryos were not affected by number of embryos, number of corpore lutea and uterine length. The uterine length was not different between on Day 6 and on Day 15. [Conclusion] The migration of porcine embryos occurs between Day 8 and Day 15 of pregnancy. Unilateral ovariotomy and ovariotomy do not affect embryonic growth and migration.

  • Isolation and Culture of Fetal Fibroblasts of Tibetan Yellow Cattle

    [Objective][Objective] To isolate fetal fibroblasts from Tibetan yellow cattle and thus to provide the ideal biological material for cloning and cellular biology of animals inhabiting in the Tibet Plateau. [Method] The fibroblast cells were isolated from tissues of Tibetan yellow fetus by explant culture method. The subculture, morphology and dynamics were observed. [Result] The fetal fibroblasts of Tibetan yellow cattle had typical morphology of fibroblast cells. The isolated cells had three growth stages including retention period (0-2 d), logarithmic phase (2-7 d) and platform phase (7-8 d). Its doubling time was 60 h. [Conclusion] Fetal fibroblasts of Tibetan yellow cattle have been successfully cultured.

  • Relevance between BMPR-IB Genotypes and Litter Size in Small Tail Han Sheep

    [Objective] To detect the polymorphism of BMPR-IB gene and analyze the relevance between genotype frequency of FecB gene and litter size. [Method] Mutation sites in the BMPR-IB gene of Small Tail Han sheep were analyzed by PCR-RFLP. [Result] The percentages of individuals with BB, B+ and ++ genotypes were 24.69%, 54.89% and 20.42%, respectively, and their average litter size per parity was 3.01, 2.81 and 2.16, respectively. [Conclusion] The BMPR-IB gene can be used as a molecular genetic marker in fecundity traits. The FecB gene can effectively increase litter size of the Small Tail Han sheep and has additive action on litter size.

  • Comparison on Egg Quality between Shandong Local Hens and Hyline Brown Hens

    [Objective] To observe differences in egg quality between Shandong local hens and Hyline brown hens. [Method] Hyline brown hens and three Shandong local chicken breeds including Jining Bairi chicken, Wenshang Luhua chicken and Laiwu black chicken were selected for egg quality comparison. [Result] The percentage of yolk, egg-shape index and shell strength were significantly higher in the Shandong local hens than in Hyline brown hens (P < 0.05), but the opposite results were found for average egg size and Haugh unit (P < 0.05). The contents of essential amino acids and linoleic acid were significantly higher in the Shandong local hens than in Hyline brown hens (P < 0.05), but no significant difference in VA content, VE content, total amino acid content and fatty acid content was found between the Shandong local hens than in Hyline brown hens. [Conclusion] The study investigated the effects of chicken breeds on egg quality, which provides a reference for improvement of chicken breeds in future.

  • Exploration on Suitable Stocking Period for Aquaculture

    To determine suitable stocking period is extremely important for increasing yield, improving quality and especially increasing breeding efficiency. The purpose of this study was to provide a technical approach for determination of suitable stocking period in aquaculture. Agricultural meteorology, mathematical statistics, methodology of economics, methodology of management, and other methods were used to explore suitability of stocking period in aquaculture. As learned from stocking of Penaeus orientalis in coastal area of Yancheng City of Jiangsu, we believed that to establish model for benefit assessment was one of ideal ways to determine suitable stocking period. In order to determine stocking period more scientifically, stocking test must be performed for specific aquatic species. The effects of environmental conditions on growth and development of juvenile aquatic livestock should be studied to identify key restraining factors, e.g., water temperature, for growth and development. The study provides the basis for establishing model for determination of suitable stocking period.

  • Prokaryotic Expression of Gene Encoding Glutamate Dehydrogenase of Streptococcus suis Serotype 2 and Preparation of Polyclonal Antibodies against Its Expressed Products

    [Objective] To obtain detection antigen for diagnosis of Streptococcus suis infection. [Method] The complete ORF of glutamate dehydrogenase (GDH) gene was amplified from the genomic DNA of Streptococcus suis serotype 2 strain SC22 isolated in Sichuan Province by polymerase chain reaction (PCR). The resulting product was cloned into the prokaryotic expression vector pET-30a, which was then transformed into E. coli BL21 (DE3). The identified positive transformants were screened for expression induced by IPTG. The expression products were subjected to SDS-PAGE and the recombinant protein was purified by nickel ion-agarose affinity chromatography. New Zealand rabbits were immunized with the purified recombinant GDH protein to prepare polyclonal antibodies. Titers of the anti-serum were determined by indirect ELISA and Western blot assay. [Result] The recombinant GDH protein was effectively expressed in the host bacteria, and highly pure recombinant protein was obtained by nickel ion-agarose affinity chromatography. High-titer anti-serum against the recombinant protein was obtained. As evidenced by western blot assay, the sera could react specifically with the lysates of all detected Streptococcus suis strains. In addition, the recombinant GDH protein could react specifically with serum samples collected from five pigs experimentally infected by strain SC22. [Conclusion] The expressed GDH fusion protein has some common epitopes of natural GDH and can be used as detection antigen to develop ELISA and other diagnostic methods.

  • Cloning and Sequencing of cSZ1 Gene of Eimeria acervulina Changchun Strain

    [Objective] To analyze Eimeria acervulina cSZ1 gene sequence and thus to find a candidate antigen for vaccine development. [Method] According to the cSZ1 gene sequence of Eimeria acervulina published in GenBank, a pair of specific primers was designed. Then, the cSZ1 gene of Eimeria acervulina Changchun strain was amplified by reverse-transcription polymerase chain reaction (RT-PCR) and sequenced. [Result] Compared with the published cSZ1 gene sequence, five nucleotide mutation sites appeared in the cSZ1 gene of Eimeria acervulina Changchun strain. They had a nucleotide sequence similarity of 99.47%, and their encoding region had a nucleotide sequence similarity of 99.61%. Among these mutation sites, two appeared in cSZ1 gene ORF, and A185G mutation led to substitution of valine by isoleucine. [Conclusion] Protein encoded by cSZ1 gene can be used as a candidate antigen for vaccine development.

  • Comparison on Immune Efficacy of Avian Influenza Virus Re-5 Strain and SS Strain in Guangxi Sanhuang Chickens

    [Objective] To compare immune efficacy of avian influenza virus (AIV) Re-5 strain (H5N1 subtype) and SS strain (H9 subtype) in Guangxi Sanhuang chickens. [Method] A total of 180 Sanhuang chickens were selected from a breeding chicken farm in Guilin City of Guangxi and assigned into four groups (n = 45). At 145 days old, the chickens in the group I and II were respectively inoculated with AIV Re-5 strain and AIV SS strain, but they were kept in cages separately. The two control groups were not inoculated. Antibody levels were determined at 170 days old. [Result] The level of antibodies against AIV Re-5 strain was significantly higher in the chickens of the group I and group II than in the corresponding control groups, respectively (P < 0.01). The level of antibodies against AIV SS strain was not significantly different between the group I and its control group or between the group II and its control group. [Conclusion] The antibody level of chickens can be increased by the AIV Re-5 strain but not by the AIV SS strain.

  • An Immunochromatographic Strip Test for Rapid Detection of Antibodies against Porcine Circovirus Type 2

    Porcine circovirus type 2 (PCV2) infection causes huge economic losses, but no serological method is available for batch detection of field samples. The aim of the study was to develop a method for large-batch detection of PCV2 infection. Colloidal gold-labeled staphylococcal protein A (SPA) was sprayed on glass fibers to prepare a conjugate pad. The recombinant ORF2 protein of PCV2 was blotted on the test line of the nitrocellulose (NC) membrane, and pig IgG was streaked on the control line of the NC membrane. Then, the immunochromatographic strip was used for detection of antibodies against PCV2. The results show that the strip test was simple and the results could be determined within 10 min with naked eyes. The test strip was highly specific for pig serum against PCV2 and no cross-reaction with pig serum against other pathogens was observed. The test strip had close similarity with ELISA. Storage at RT for 6 months did not affect the specificity and sensitivity obviously. A total of 324 clinical pig sera were detected by both ELISA and the developed test strip, and the coincidence was 98.77%. Therefore, the developed immunochromatographic strip is specific, sensitive, stable, fast and simple, and it is suitable for on-site detection of antibodies against PCV2.

  • Effect of Copper Excess on Lymphoid Organs in Broilers

    The experiment was conducted to examine the effect of dietary copper excess on lymphoid organs by the methods of experimental pathology and flow cytometry (FCM). A total of 350 one-day-old avian broilers were divided into seven groups and fed on diets as follows: 1. control group (Copper 11 mg/kg·diet), 2. Copper excess (Copper 100 mg/kg, copper excess group I; copper 200 mg/kg, copper excess group II; copper 300 mg/kg, copper excess group III; copper 400 mg/kg, copper excess group IV; copper 500 mg/kg, copper excess group V; copper 600 mg/kg, Cu excess group VI) for 6 wk. The weight and growth index of the thymus, spleen and bursa of Fabricius were obviously decreased (P < 0.05 or P < 0.01) in the copper excess groups III, IV, V and VI, and were lightly increased in both the copper excess group I and copper excess group II when compared with those of the control group. The G0/G1 phase of the cell cycles of the aforementioned lymphoid organs was higher (P < 0.05 or P < 0.01), and S, G2+M phases and proliferating index lower (P < 0.05 or P < 0.01) in the copper excess groups III, IV, V and VI than in the control group, copper excess group I and copper excess group II. At the same time, it was also observed that the number of lymphocytes was decreased in the lymphoid organs in the copper excess groups III, IV, V and VI. The results demonstrated that dietary copper in excess of 200 mg/kg·diet impaired the progression of lymphocytes from G0/G1 phase to S phase obviously, inhibited the development of lymphoid organs and caused pathological changes in the lymphoid organs, and that copper of 100 and 200 mg/kg·diet was helpful for the development of the lymphoid organs. Potential mechanisms underlying the aforementioned observations are discussed.

  • Efficacy of Chinese Herbal Medicine against Cow Endometritis

    [Objective] To investigate safety and clinical efficacy of compound Chinese herbal medicine (Gongshukang) against cow endometritis. [Method] Clinical trials were constructed in cow community of Qingzhen and Wudang. A total of 311 parental cows suffering from endometritis were selected and randomly divided into two groups. The cows in the group I were gavaged with Gongshukang, and those in the group II were administrated with long-acting oxytetracycline hydrochloride. Therapy was based on the severity of illness. [Result] The cure rate and effective rate were high in mild and moderate cases of the two groups, but the cows in the group I needed shorter time to be in heat than those in the group II. The pregnancy rate was significantly higher in the group I than in the group II (P < 0.05). For severe cases, the cure rate of the group I was significantly lower than that in the group II (P < 0.01), but the effective rate of the group I was significantly higher than that in the group II (P < 0.05). [Conclusion] The Gongshukang has antibacterial activity and anti-inflammatory activity, induces sexual desire and increases pregnancy rate. It has better efficacy against mild and moderate endometritis than long-acting oxytetracycline hydrochloride, but its efficacy is worse in severe cases.

  • Determination of Gastrodin Content in Gastrodia elata in Bomi of China by High-Performance Liquid Chromatography

    [Objective] To select excellent varieties of Gastrodia elata and to determine the most suitable picking season of the plant. [Method] The gastrodin contents in fresh wild G. elata harvested in winter in Bomi County, artificially cultivated fresh G. elata harvested in winter in Bomi County, and nonlocal G. elata were determined by high-performance liquid chromatography (HPLC) under the following conditions: chromatographic column, AngilentTc-C18 (5 μm, 4.6 mm × 250.0 mm); mobile phase, acetonitrile-water (5∶95, V/V); wavelength, 220 nm; flow rate, 1.0 ml/min; column temperature, 35°C; and injection volume, 10 μl. The gastrodin contents in G. elata respectively harvested in winter and summer were compared. [Result] The gastrodin content was higher in the fresh wild G. elata harvested in winter in Bomi County (0.076%) than in the artificially cultivated fresh G. elata harvested in winter in Bomi County (0.045%) and nonlocal G. elata (0.025%). Additionally, the gastrodin content was higher in G. elata harvested in winter than in those harvested in summer. [Conclusion] G. elata from Bomi Country is an excellent variety and should be harvested in winter.

  • Detection of Salmonella in Forage by Dot-ELISA

    [Objective] To develop a dot-ELISA (dot-enzyme linked immunosorbent assay) method for detection of Salmonella in forage. [Method] Forage samples including 100 finished feed samples, 85 fishmeal samples and 50 protamine samples were randomly collected from a feed factory in Xining City, and Salmonella in these samples were detected by dot-ELISA. The Salmonella isolates were further identified using biochemical tests and serological tests. [Result] As detected by dot-ELISA, the positive rates of Salmonella in the finished feed, fishmeal and protamine were 38.00%, 62.35% and 60.00%, respectively. The coincidence rate between the dot-ELISA and routine identification method was 82.23%. [Conclusion] The dot-ELISA has good specificity and sensitivity and is highly coincidental with the routine identification method. Therefore, dot-ELISA can be used for detection of Salmonella in forage.

  • Effect of Fertilization on Growth and Development of Red Clover

    [Objective] To investigate the effects of fertilization on growth and development of red clover. [Method] American red clover "Agivasion" was used as tested materials. In field, four treatments were used, including no fertilization (CK), application with N fertilizer, application with NP fertilizer, and application with NPK fertilizer. And the effects of fertilization on growth and development of red clover were analyzed. [Result] After fertilization, plant height was increased, particularly in application with NP fertilizer. Branching was promoted, particularly in application with NPK fertilizer. Rhizome became thicker, particularly in application with NP or NPK fertilizer. The fertilization promoted accumulation of dry matter in stems and leaves and increased yield per plant significantly at the 0.01 level. This promoting effect became weaker in such order: NPK fertilizer > NP fertilizer ≈ N fertilizer > CK. [Conclusion] Rational fertilization can promote growth and development of red clover.
