• Molecular Cloning and Sequence Analysis of Mitochondrial DNA ATPase8 gene in Guyuan Native Chicken

    [Objective] To amplify ATPase8 genes from genomic DNA of Guyuan chicken (white plume, ma plume and red plume), Arbor acres broiler and Roman layer. [Method] Using specific primers for differential detection of chicken-derived ingredients, the ATPase8 genes were amplified from the genomic DNA extracted from chicken blood by PCR. And the PCR products were sequenced. [Result] The full length sequence of chicken ATPase8 was 165 bp in the Guyuan chicken and Arbor acres broiler and 168 bp in Roman layer. [Conclusion] The ATPase8 gene of the Guyuan chicken has high homology to that of the Arbor acres broiler or Roman layer, but it has low homology to that of quail, partridge, ostrich, goose or turkey.

  • Polymorphism of Coding Sequences of IGF1R Gene in Baise Horses and Thoroughbred

    [Objective] The aim was to study polymorphism of mat-peptide sequence of IGF1R gene in Baise horses and thoroughbred,which would afford reference for the further studies on the dwarf mechanism and molecular breeding in horses.[Method] A total of 57 blood samples of each breed were collected and genomic DNA was extracted by the standard phenol-chloroform method.Five DNA pools of each breed were constituted and polymorphism sites were identified by sequencing PCR products.Frequencies of genotypes and alleles at these sites of each breed were checked by PCR-RFLP.[Result] Four polymorphism sites were identified in exon 2,5 and 16,including mutations of T406C,T179 627C,G212 077A and G212 110A.No difference was found in the frequency of T179 627C between the Baise horses and thoroughbred.The mutation G212 077A was only found in the thoroughbred,and the mutations,T406C and G212 110A,were only checked out in the Baise horses.[Conclusion] Whether these mutations are associated with horse growth needs further studies.

  • Breeding of Silkworm Variety "Chuancan No.24" with Strong Vitality for Summer-Autumn Rearing

    [Objective] The aim was to introduce the breeding process of natural colorful cocoon silkworm variety,Chuancan No.24.[Method] By cross breeding and pedigree separation,the silkworm variety,Chuancan No.24,with double sex-limited markings and strong vitality and for summer-Autumn rearing was bred.[Result] The results of laboratory identification and countryside test in Sichuan Province indicated that the Chuancan No.24 was strong and could be reared easily.They had uniform development and excellent quality of cocoon and silk.In addition,their sexes could be identified by larval body color and markings.The main indexes are as follows:cocoon yield per 10 000 fourth instar larvae,19.66 kg;filament length,1 241.29 m;length of non-broken filament,941.99 mm;reelability,75.87%;raw silk ratio of fresh cocoon,17.07%;and neatness,94.03 points.The main economic features of new variety were better than those of the control.[Conclusion] The Chuancan No.24 has been approved by the Sichuan Silkworm Evaluation Commission,and it is suitable to rear this new variety in some regions of the Yangtze River drainage basin,such as Sichuan Province and Chongqing.

  • Effect of Different Oligosaccharides on Immunity and Production Performance of Piglets

    [Objective] To study the effects of different oligosaccharides on immunity and production performance of piglets.[Method] A total of 75 Yorkshire×Rongchang piglets were assigned into five groups,15 in each group.Isomalto-oligosaccharide (IMO),fructo-oligosaccharide (FOS),mannan-oligosaccharide (MOS),and mixed oligosaccharide were supplemented to basal diet (7.5 g/kg),respectively.The control group was also set.The experimental period lasted for 53 d.The clinic symptoms and diarrhea were observed.All piglets were weighed once a week.Feed intake was also recorded.Blood was collected via precaval vein for determination of immunology indexes at the age of 30 and 60 d,respectively.[Result] The IMO and FOS enhanced cellular immunity of 30-day-old piglets significantly (P<0.05),and the IMO also enhanced their humoral immunity.The MOS enhanced cellular immunity and humoral immunity of 60-day-old piglets significantly (P<0.05).It also improved production performance largest,and its diarrhea rate was the lowest.[Conclusion] Different oligosaccharides have various effects on immunity and production performance of piglets.

  • Detection of Telomerase Activity and Expression of TERT Gene in Antler of Sika Deer

    [Objective] To observe the expression of telomerase in antler of sika deer during growth stage and thus to provide a new line for revealing the mechanisms of development and regeneration of antlers.[Method] The updated TRAP (Telomeric repeat amplification protocol) argentum-dying method was used to detect the activity of telomerase in mesenchyme,precartilage,cartilage and ossification tissue of antler tip during the growth stage.Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of telomerase reverse transcriptase (TERT) in these cells.[Result] The activity of telomerase was detected in the mesenchyme,precartilage and cartilage but not in the sclerotic tissue.The relative telomerase activity in the mesenchyme,precartilage and cartilage was 91.2,46.4 and 13.7,respectively.In addition,the results of the expression of TERT gene also supported the detection result of telomerase activity.[Conclusion] During growth phase,telomerase is expressed in multiplication area of deer antlers,and its activity decreases gradually with further differentiation of cells in antlers.Therefore,telomerase may play a key role in development and regeneration of antlers.

  • Dynamic Changes of Serum Leptin and Ghrelin and Their Correlation in Buffalos around Estrus

    [Objective] To understand variation and correlation of serum leptin and ghrelin in buffalos around estrus.[Method] From Day 14 before estrus to Day 22 after estrus,blood was collected from 23 Murrah buffalos in a Guangxi buffalo farm.Serum levels of leptin and ghrelin were determined by ELISA.[Result] The serum levels of leptin and ghrelin increased steadily before estrus but changed greatly in estrus.They had significantly positive correlation (P<0.01).[Conclusion] This study provides a new way for research about mechanism of nutritional and metabolic diseases in buffalos.

  • In Vitro Bacteriostatic Activity of Five Chinese Herbal Medicines against Two Common Pathogens of Scylla serrata

    [Objective] To investigate the in vitro bacteriostatic activity of Chinese herbal medicines (rhubarb,Baikal skullcap root,honeysuckle,eucommia bark and Chinese date) and their compounds against two common pathogens of Scylla serrata (Vibrio parahaemolyticus and Aeromonas hydrophila).[Method] Drug sensitivity of the bacteria was determined by paper method.Minimum inhibitory concentration (MIC) andminimum bactericidal concentration (MBC) were determined by dilution method and plate method,respectively.[Result] The bacteriostatic order of single medicines on V.parahaemolyticus were Baikal skullcap root>honeysuckle>rhubarb>Chinese date>eucommia bark.The bacteriostatic order of the compounds were A=B=C>D.Among these medicines,the Baikal skullcap root and the compounds A,B,C and D had better bactericidal effects on V.parahaemolyticus.The bacteriostatic order of single medicines on A.hydrophila was rhubarb>Baikal skullcap root>honeysuckle>Chinese date>eucommia bark.The bacteriostatic order of the compounds were A>B>C=D,while the rhubarb and the compound C had better bactericidal effects on A.hydrophila.[Conclusion] The Chinese herbal medicines including rhubarb,Baikal skullcap root,honeysuckle,eucommia bark and Chinese date have good bacteriostatic effects on V.parahaemolyticus and A.hydrophila,and they can be used to treat bacterial infection in Scylla serrata.

  • Development of Multiplex-PCR for Identification of Pasteurella multocida,Haemophilus parasuis and Actinbacillus pleuropneumoniae

    [Objective] To develop multiplex-PCR for diagnosis of mixed infection caused by Pasteurella multocida (PM),Haemophilus parasuis (HPS) and Actinbacillus pleuropneumoniae (App).[Method] PCR method was developed to detect single infection caused by PM,HPS or App.The conditions of amplification and primers were optimized,and the multiple-PCR was developed to detect mixed infection of PM,HPS and App.[Result] A 457-bp band,a 821-bp band and a 342-bp band were simultaneously amplified in the one PCR reaction system.The method had high sensitivity and specificity.[Conclusion] The multiple-PCR is successfully developed and can be used for differential diagnosis of PM,HPS and App.

  • Comparison of Two Construction Methods of Targeting Gene of Haemophilus parasuis

    Haemophilus parasuis is one kind of constant bacteria in porcine upper respiratory tract,and it can cause multiple serositis,arthritis and other diseases under certain conditions.Due to lack of efficient genetic operating system,its pathogenic mechanism is not very clear.Ligation with DNA ligase and fusion PCR were used to construct targeting hhdA gene of Haemophilus parasuis,respectively.The fidelity,application scope,operation and conditions of the constructed fusion fragments were compared.The results showed that construction with DNA ligase was more mature technology as manifested by more stable conditions and more extensive application.The fusion PCR method had high fidelity and simple operation,and the transformation rate was 9.5 times as high as that of ligation with DNA ligase.For this reason,this method was more suitable for construction of multi-fragment targeting genes.The study lays a foundation for establishing an efficient operating system of targeting gene of Haemophilus parasuis in the future.

  • In Vitro Activity and Postantibiotic Effect of Tulathromycin against Actinobacillus pleuropneumonia

    The postantibiotic effect (PAE) is defined as the suppression of bacterial growth persisting after a short exposure of bacterial cultures to an antibiotic.This phenomenon has obvious clinical interest,because antibiotic agents which produce extensive PAEs could be administrated less frequently without reducing efficacy.Theminimum inhibitory concentration (MIC) and PAE of tulathromycin and erythromycin on five isolates of Actinobacillus pleuropneumonia were determined.All strains were susceptible to tulathromycin (MIC:0.25-1.00μg/ml) and resistant to erythromycin (MIC:32-128μg/ml).The mean PAEs of 1×MIC,2×MIC,4×MIC and 8×MIC of tulathromycin against the tested isolates were 0.62,1.53,2.70 and 4.50,respectively.These results indicated that tulathromycin might be administered at longer time intervals than those already applied.This is in favorite of the clinical rational use of antimicrobial drugs.

  • Diagnosis of Mixed Infection of Classical Swine Fever Virus and Porcine Reproductive and Respiratory Syndrome Virus and Pathogenic Characteristics

    [Objective] To diagnose swine diseases caused by CSFV (Classical swine fever virus),PRRSV (Porcine reproductive and respiratory syndrome virus) and PRV (Pseudo-rabies virus) and analyze the pathogenic characteristics.[Method] The tissues and viscera of the diseased swine were collected from a hoggery in Fujian Province.DNA and RNA were extracted for PCR amplification and sequencing.ELISA method was used to determine CSFV,PRRSV and PRV infection.[Result] The sequencing analysis and ELISA results showed that the mixed infection was caused by CSFV,PRRSV and PRV.[Conclusion] The swine epidemic situation was mainly caused by CSFV and PRRSV.

  • Isolation,Identification and Pathogenic Characteristics of Duck Hepatitis Virus Isolates

    [Objective] The aim of this study was to determine and analyze the pathogen and the reasons of duck viral hepatitis which is prevalent recently and difficult to control.[Method] Viruses were isolated from livers and spleens of ducks with typical clinical symptoms in Linyi,Weifang,Binzhou and other regions of Shandong Province.The pathogenic characteristics were observed by inoculation in chicken or duck embryo,RT-PCR,serological test,and duck regression.[Result] Four duck hepatitis virus (DHV) strains were isolated,and the 5th passage allantoic fluid contained 103.41-105.20 ELD50/ml.The serum cross protection rate was 20%-80% between the DHV stains and DHV type I.The mortalities of 4-day-old healthy ducks challenged by these four stains were 50%-100%.All challenged ducks had typical lesions of duck viral hepatitis,and the death peak appeared after 24-48 h.[Conclusion] The virulence of different DHV isolates has regional difference.

  • Comparison on Infectious Bursal Disease Monoclonal Antibodies Prepared with Two Different Immunogens

    [Objective] To compare the characteristics of monoclonal antibodies against infectious bursal disease virus (IBDV) prepared with two different immunogens,VP2 protein expressed by prokaryotic system and purified IBDV.[Method] IBDV VP2 gene was amplified by RT-PCR and expressed in a prokaryotic system.The recombinant protein was purified by affinity chromatography.IBDV was purified by ultracentrifugation.Balb/c mice were immunized with the purified recombinant protein and IBDV,respectively.The monoclonal antibodies were screened by ELISA.[Result] Two cell lines secreting antibodies against IBDV VP2 protein were obtained,and their ELISA titers were 1:2×104.Four cell lines secreting antibodies against IBDV were produced,and their ELISA titers were 1:2×106,1:6×104,1:1×105 and 1:4×103,respectively.All monoclonal antibodies specifically bound to their own immunogen but did not react with other viruses or proteins.After 10-20 passages,these cell lines still secreted antibodies stably.[Conclusion] The monoclonal antibodies prepared with the recombinant IBDV VP2 protein or purified IBDV can induce immune response in mice,and VP2 specific monoclonal antibodies can be obtained with VP2 protein expressed in the prokaryotic system as immunogen.

  • Mutagenic Effects of Spaceflight Treatment on the Second-Generation Seeds of Alfalfa

    [Objective]To explore the effects of spaceflight on the second-generation seeds of alfalfa and provide a theoretical basis for mutation breeding.[Method]The seeds of Medicago stavia L.lines no.1,no.2 and no.4 were carried into space by the Shijian-8 seed breeding satellite for a 15-d spaceflight treatment.After returning to the ground,seedlings were transplanted to field.Traits of the second-generation seeds of alfalfa were evaluated.[Result] The 1 000-grain weight of the second-generation seeds were 5%-9% significantly higher than that the control (P<0.05).The germination rate,seedling weight,shoot length and root length were significantly increased (P<0.05).The hard seed rate and the rate of moldy seeds were significantly decreased (P<0.05).However,the rate of dead seeds was increased.[Conclusion]Spaceflight treatment has positive mutagenic effects on the second-generation seeds of alfalfa.
